Leserbrief zu Bremslichtern an E-Autos:

Das Problem tritt (ggf. - Erklrung weiter unten) auch in der EU auf. Zunchst die rechtliche Seite: Bremsleuchten sind in D in 53 StVZO geregelt. Neben vorgeschriebener Anzahl, Ortder Anbringung etc. steht da dann auch in Absatz 2: "Die Bremsleuchten drfen auch bei Bettigen eines Retarders oder einer hnlichen Einrichtung aufleuchten.
Man beachte: drfen, mssen tun sie nicht.

Kommen wir zu den e-Autos. Ich habe da gerade zwei in Betrieb, einen lteren e-Corsa und einen relativ neuen Kia.
Beim Corsa gibt es 3+1 Schaltstellung (vulgo Gnge) P(ark), N(eutral), D(rive) und eben den B(ergabmodus). Der Modus Bhat zu D den einzigen Unterschied, dass der Motor strker rekuperiert, ergo das Auto deutlich strker abbremst wenn man den Fuss vom Gaspedal nimmt. In diesem Fall leuchtet beim Opel das Bremslicht NICHT, obwohl das Auto fr den restlichen Verkehr berpropertional langsamer wird. Im D-Modus rekuperiert der Opel eh nicht wesentlich anders als ein Verbrenner beim Gas-Wegnehmen langsamer wird.

Der Kia wiederum hat nur die drei Fahrstufen P, N und D. Dafr erlaubt er dem Fahrer, die Rekuperation in vier Stufen selbst einzustellen. Das geht von Ich fahre wie ein Verbrenner mit fast keiner Rekuperation bis hin zum Modus Autoscooter, in dem das Auto nur mit dem Gaspedal gefahren werden kann und ziemlich heftig bremst wenn man den Fuss vom Gas nimmt. Nur im ersten dieser Modi leuchtet das Bremslicht beim Rekuperieren nicht, in allen anderen schon.

Meine Vermutung ist, dass die Hersteller durchaus das Gefahrenpotenzial einer nicht angezeigten Brutal-Rekuperation erkannt haben und da selbststndig aktiv geworden sind. In einem anderen Forum habe ich aber noch diese Aussage gefunden: "Das Gesetz schreibt vor, wenn die Bremsanlage (egal ob Retarder, Motorstaubremse, ACC...) mehr als 0,7m/s verzgert kann das Bremslicht aktiviert werden, ab 1,3m/s muss das Bremslicht aktiviert werden. Da konnte ich aber keine Verschrift oder Gesetz zu finden.

Die Werte machen aber Sinn, da bis zu 0,7 m/s ungefhr das ist, was ein Auto beim Gas-Wegnehmen auch verzgert. Sollte es so eine Vorschrift geben, dann wrde ich weiter vermuten, dass z.B. der B-Modus im Opel nur bis max 1,3 m/s verzgert und somit in den optionalen Bereich fllt. Gemessen habe ich das aber nicht ;) Spass am Rande: seit dem letzten Firmware-Update des Opels ist die Rekuperation im B-Modus deutlich schwcher geworden. Evtl. gibt es also tatschlich zumindest eine EU-Vorgabe und da man das Bremslicht nicht steuern konnte hat man bei Stellantis das Problem halt ber eine reduzierte Rekuperation via Software gelst.

Fazit: Zumindest bei den neueren, mir bekannten e-Autos leuchten die Bremslichter bei Einsatz von starker Rekuperation, bei lteren....

In a recent podcast appearance, Arthur Hayes, the former head of BitMEX, delivered a sobering prediction about the largest cryptocurrency by market capitalization, Bitcoin (BTC).

Hayes believes that Bitcoins anticipated climb to an all-time high of $70,000 is unlikely to materialize this year, dampening the hopes of investors and traders. However, he points to a potentially bullish turn in 2024, driven by a significant event that could shape the markets trajectory.

Bitcoin Rise To $70,000 To Be Delayed

During an episode of the What Bitcoin Did podcast, Hayes shared his perspective on Bitcoins price trajectory. He indicated that breaching the $70,000 milestone is not expected to occur in 2023.

Instead, Hayes suggests that next years anticipated halving event will play a crucial role in determining the markets direction. While the delay may disappoint some, he remains optimistic about a potential breakthrough in the near future.

The former head of BitMEX believes that the upcoming 2024 halving, an event that slashes Bitcoins block reward in half, will be a pivotal moment for the market. He suggests that crossing the $70,000 barrier will likely happen during that time.

However, his long-term forecast raises concerns about an impending blow-off top in 2025 or 2026, potentially signaling a significant societal event or turmoil which he calls Armageddon. Hayes attributes this potential turmoil to factors such as excessive money printing and growing global discontent.

Hayes noted:

We have got this halving next year, 2024. I think thats gonna be a good year. I dont think we get up to $70,000 this year. Next year is when we cross that barrier and then we get the blow-off top 2025, 2026. And then its Armageddon.

BTC Latest Price Action

Meanwhile, Bitcoin has shown a possible brewing uptick in the past week. The largest crypto asset by market capitalization has surged 4.3% in the past week. Over the past 24 hours, BTC has seen a 2.8% gain.

Music may be as old as mankind, but the songs of the ancient past are largely lost to history. Among those to create and play music were the ancient residents of Mesopotamia. Within this cradle of civilization, history, laws, and religious texts were recorded in cuneiform on tablets. Many have survived, including an exceptional tablet: the earliest extant sheet music in the world. Known as Hurrian Hymn No. 6, the 3,400-year-old tablet records a tune in praise of the ancient goddess Nikkal.

The ancient Hurrians were a Bronze Age people of the Near East. They had a rich culture, although much of their history and origins remain shrouded in mystery. They left behind clues however, such as a tablet bearing cuneiform discovered in the 1950s in the ruins of the city of Ugarit in northern Syria. The city was a center of trade and exchange with other Near Eastern civilizations, and even kingdoms as far as Egypt. On the tablet are four lines of text in the Ugarit dialect. While the language is difficult to interpret, the Hurrian Hymn No. 6 is clearly the world's earliest extant melody or song.

The words of the song lie in the upper quadrant, while the lower contains the notes. Scholars believe that sections of repeated lyrics indicate a refrain. Yesterday, Polkadot (DOT) reached its weekly high at $5.54 but subsequently started a corrective phase. In the past 24 hours, DOT has managed to retain 1.8% of its overall gains during that period. On the weekly chart, DOT exhibited a 3% price increase.

However, the technical analysis suggests that bears currently have the upper hand, with buying strength and demand remaining low. Despite this, if Bitcoin continues its upward movement toward the $28,000 level, several altcoins, including DOT, may also experience positive price action.

The overhead resistance for DOT is currently positioned at $5.58. If the altcoin manages to surpass this resistance level, it could potentially reach $5.80. Despite the price correction, DOT has thus far maintained a position above the crucial support level of $5.40.

However, if sellers gain momentum, the price may drop below the vital support level of $5.40 and trade around $5.30. The trading volume of DOT in the previous session was bearish, indicating a decline in demand driven by the bears on the daily chart.

I haven't spent much time researching this topic yet, but I hope that some of you can point me in a direction.

I've been reading about the processed food industry and it makes me feel thankful that I'm cleaning up my nutrition and eating more whole foods.

I have a feeling that the products we buy for skincare, sunscreen, and insect repellent may be equally chemical ridden and awful for us.

A joint population-based retrospective cohort study from researchers in Canada and Israel has found that inducing labor through oxytocin administration does not increase the risk of autism spectrum disorder (ASD) in offspring. The current studys design examined nine different exposure groups from the two countries that were not mutually exclusive. In Canada, 170,013 (41.0%) of the cohorts deliveries were not induced or augmented, 107,543 (26.0%) were exposed to oxytocin during induction and/or augmentation, and 136,780 (33.0%) were induced or augmented but not exposed to oxytocin. In Israel, 51,790 (62.5%) of the cohorts deliveries were not induced or augmented, 28,852 (34.8%) were exposed to oxytocin during induction and/or augmentation, and 2,250 (2.7%) were induced or augmented but not exposed to oxytocin. The participants were followed for a mean of 8.43 years in the Canadian cohort and 4.33 years in the Israeli cohort to determine if an autism diagnosis was given. After analyzing the data and adjusting for maternal sociodemographic characteristics, pregnancy conditions and risk factors, labor and delivery characteristics and neonatal characteristics, the authors concluded that no associations were found in their primary analysis between oxytocin induction and the risk of ASD in offspring. This finding contradicts a 2016 study that identified a higher risk of autism in infants born to mothers who received pitocin (synthetic oxytocin) during labor. The authors of this current study suggest that the mother or infants underlying health issues were not considered and likely confounded previous research, linking autism to oxytocin use to induce labor.

Preterm Infants Encounter Toxic Metals During Their Stay in the NICU "IndyWatch Feed Allcommunity"

Total Parenteral Nutrition and Red Blood Cell Transfusion Provide Exposure Route

Premature infants frequently stay in a neonatal intensive care unit (NICU) for specialized medical care. Total parenteral nutrition (TPN) and red blood cell (RBC) transfusions are two types of clinical management and specialized care that premature infants with low birthweight encounter in the NICU. A recent study has assessed the exposure of premature infants to toxic metals during their stay in the NICU from TPN and RBC transfusions and the potential risk of neurodevelopmental delay at age 2 months. The research included 33 preterm infants that required TPN and RBC transfusions. Blood samples from the infants were collected for metal analysis at two different time points (admission and before discharge). Metals in the daily TPN were analyzed, and neurodevelopment was assessed using the Ages and Stages Questionnaire Edition 3 (ASQ-3). Shockingly, the authors discovered that all samples of TPN had metal contamination: 96% exceeded the critical arsenic limit (0.3 g/kg body weight/day). They also found that the added daily manganese intake from TPN for preterm infants surpassed the recommended dose (1 g/kg body weight), raising safety concerns. Additionally, the authors discovered that all samples of RBC transfusions exceeded the estimated intravenous reference dose for lead (0.19 g/kg body weight). Levels of mercury, lead and manganese in preterm infants at discharge decreased 0.867 g/L (95% CI, 0.76, 0.988), 0.831 (95%CI, 0.779, 0.886) and 0.847 g/L (95% CI, 0.775, 0.926), respectively. The researchers identified a decrease in ASQ-3-problem solving scores associated with higher levels of blood lead in preterm infants taken at admission ( = 0.405, 95%CI = 0.655, 0.014) and with plasma manganese ( = 0.562, 95%CI = 0.995, 0.172). They also observed an association between decreased personal social domain scores with higher blood lead levels of preterm infants before discharge ( = 0.537, 95%CI = 0.90...

Bitcoin on-chain data suggests a historical support line may have helped the coin once again as the asset has recovered toward $28,000 today.

Bitcoin Short-Term Holder Cost Basis May Still Be Active As Support

According to data from the on-chain analytics firm Glassnode, the BTC price approached the cost basis of the short-term holders recently. The relevant indicator here is the realized price, which is a metric derived from the realized cap.

The realized cap refers to a capitalization model for Bitcoin that says that the value of each coin in the circulating supply is not the current spot price, but the price at which it was last transacted on the blockchain.

In this way, the model accounts for the price at which each investor acquired their coins. That is, their cost basis. When the realized cap is divided by the number of coins in circulation (to find a sort of average value), the aforementioned realized price emerges.

This realized price signifies the average value at which each holder in the market bought their coins. The metric can also be defined for only partial segments of the market, like the short-term holders (STHs), in which case, the indicator will tell us about the average cost basis among this group only.

The STHs are all those investors who bought their coins less than 155 days ago. The BTC holders outside this group are termed the long-term holders (LTHs).

Now, here is a chart that shows the trend in the Bitcoin realized price for the STHs over the past couple of years:

The fight is on to hold the United States government responsible for colluding with social media companies to censor Americans free speech rights online.

Missouri v. Biden, which was filed on May 5, 2022, has been taking quite the trip through the court system. It was amended three separate times, most recently to add an amendment that transforms the case into a class action suit due to the sheer number of Americans impacted by the governments crimes.

Uncover DC has been tracking the case, offering play-by-play details about what has been happening with the case over the past year. The plaintiffs, including the states of Missouri and Louisiana, pushed for expedited discovery to obtain a limited set of evidence and depositions from certain individuals.

They argued that this evidence would allow them to make the case for a temporary injunction to stop the government from infringing on the first amendment rights of Plaintiffs and their citizens, Uncover DC reported.

The judge granted the motion for expedited discovery and depositions, prompting a fight between the government and the judge, in this case, Judge Terry Doughty. In short, the defendants want to stop all discovery and certain plaintiffs from being deposed.

Fortunately for the plaintiffs, Judge Doughty disagreed, forcing the CISA head, White House Press Secretary Jen Psaki, and other alleged co-conspirators to sit down and tell all about what they did to deprive Americans of their First Amendment rights.

Psaki, as you may recall, made threats to social media companies straight from the podium, which prompted her to be deposed. She then left her White House position, conveniently.

It is set in a fictional world gripped by perpetual war between a sea-faring economic power and a population of monsters who appear to attack ships out of nowhere. The hunters, the heroes of the story, patrol the seas in a near-constant state of combat, fighting to keep trade routes open and safe. It was a fairly overt critique of the military industrial complex, though the deeper theme that I saw was the toxic effects that the media propaganda complex has on a society, reducing peoples ability to exercise discernment, and obscuring objective truth.

I think the reason why The Sea Beast had such an impact on me is because at its core, it is a story of what can happen to society when truth becomes obscured by revisionist history and propaganda. In the end, it was up to individuals to exercise discernment, and break the societal trance that had been propagating violence for generations. American culture and history is rich with individualism and critical thought, such as the idea that taxation without representation is immoral. What is the TVTP?

The Targeted Violence and Terrorism Prevention Grant Program (TVTP) is administered by the Department of Homeland Security. It provides funding to funds to public, private, and non-profit institutions like colleges, universities, and county governments to establish or enhance capabilities to prevent targeted violence and terrorism.

On the face, it doesnt sound that bad.

But when you look closer, youll see that this funding is only doled out to groups that align with a specific agenda. And that agenda is to target conservatives, Christians, and Republicans in what MRC calls a coordinated effort to make America into a one-party system.

The Biden administration has awarded 80 grants through the TVTP totaling just under $40 million. The lowest grant was for $85,000, the highest was over $1.1 million, and the median was about $442,000. TVTP grant recipients are prohibited from engaging in viewpoint discrimination, according to DHS.

Started by the Obama administration under a different name, the TVTP was broadened and revamped by the Biden administration with a new focus on violent extremism and white supremacy. DHS named one of its TVTP goals as media literacy and online critical thinking initiatives, which many grantees listed as the mission of their projects.

The most I can do for him is to let God know that although he listens to devil music, and wears all black, that he doesnt deserve any vengeful wrath, hes just a confused kid. I have to admit though, the severed head in the microwave on that shirt was impressively realistic, but not as impressive as the power of prayer.

Metal expert Todd Fisher gave some examples of other scenarios where the shirt choices of metalheads got them into hot water.

You wouldnt think in this day and age anyone would get bent out of shape over a dumb t-shirt, but thats sadly not the case, Fisher explained. Coffee Company Releases Punk Album "IndyWatch Feed Allcommunity"

CHICAGO Local coffee roasters Undefeated Coffee Collective surprised its followers by deciding to release a punk album 22 years into their existence, confirmed multiple sources still waiting for their French press to finish.

Weve been doing collaborations with punk bands for years but its always a one-way street. We name a special roast after the band, like our Rancid Rancid Brew or our None More Black Coffee and we wanted to mix it up, said Bruce Smalls, co-founder of Undeefeated Coffee Collective. And frankly, if were going to have to compete with Green Days coffee, Id just as soon they compete with us and our energetic blend of punk rock. Since anyone can just do anything they want, you know?

The album, titled Grounds For a Revolution, received largely positive reviews online since its release earlier this month.

Honestly not bad for a bunch of guys that make coffee, said Ed Highsmith, an online music journalist. You could tell me this was a real band and I dont know if Id be able to tell the difference or not. True, theyre singing about coffee on every single song, but you could say the same about the Descendents and not be completely wrong. Musically, however, the bold choruses mix really well with the smooth verses. I even think I detected some notes of metal and ska at times. Grounds For a Revolution is a great album to start your day.

Established up-and-coming punk bands were not as enthusiastic about the release.

You think what we do is easy? Ive spent years perfecting the same three chords and basic progressions, over a loosely similar baseline and drumbeat, said vocalist and lead guitarist of punk band Eddies Gotta Go, Dan Harper. I bet they wouldnt be excited if I took a trip down Colombia to source coffee beans, made sure all the workers were treated fairly in the process, and then packaged and sold it here in the States at an inflated price because it has a vaguely punk aesthetic. They are lucky Im too fucking lazy to even begin to attempt something like that.

As of press time, Undefeated Coffee Collective confirmed upcoming plans to expand into brewing beer and releasing metal albums.

No one criticizes leftists more than other leftists, and ever since I told my DSA working group that I enjoy the smash hit series Yellowstone, it feels like Rose Emoji Twitter is out to crucify me. Its gotten to the point where Ive begun to question my entire identity and core beliefs; so I ask you, dear reader, can I still be a leftist if I watch Yellowstone?

I get that Yellowstone has been branded by some as anti-woke, and that its the favorite show of just about every Republican suburbanite with an F150 that will never see a job site. But liking something that conservatives have overwhelmingly aligned themselves with shouldnt call my politics into question. Right?

I still think all cops are bastards. I still think landlords are leeches. I still think billionaires shouldnt exist; especially the ones trying to build an airport on The Yellowstone Ranch. Yes, I get that it would create jobs and yes I get that no one person should own as much land as John Dutton, but I mean, come on, like anyone is going to side with Market Equities.

All the people in my DSA chapter who I used to call my comrades have dared to even question my allyship with Indigenous communities, and environmental values! While I would love to see large swaths of land returned to indigenous peoples in real life, you gotta admit, the Yellowstone writers kinda make you root for the Duttons. And yes, the cattle industry is disastrous for the environment, but aerial shots of grazing cows are beautiful and roping calves does seem kinda fun.

There are some parts of the show that we should all be able to agree are good, too. They kill a lot of white supremecists, which is undeniably cool. Sure, if the Duttons were real they would probably share some of the same beliefs to a degree, but watching them shoot up a bunch of Nazis while Sleeping On The Blacktop is playing was fucking awesome.

Look, we all like something thats at least a little problematic. Im willing to bet more than a few leftists I know still listen to Brand New. So you know what? Im not taking leftist out of any of my bios. Im the type of guy that will lead a protest outside of a store selling fur, and then enthusiastically watch cattle get branded and sold for slaughter on a fictional television show, and if you dont like that, tough.

Abstract:  Lipids play a crucial role in the entry and egress of viruses, regardless of whether they are naked or enveloped. Recent evidence shows that lipid involvement in viral infection goes much further. During replication, many viruses rearrange internal lipid membranes to create niches where they replicate and assemble. Because of the close connection between lipids and inflammation, the derangement of lipid metabolism also results in the production of inflammatory stimuli. Due to its pivotal function in the viral life cycle, lipid metabolism has become an area of intense research to understand how viruses seize lipids and to design antiviral drugs targeting lipid pathways. Palmitoylethanolamide (PEA) is a lipid-derived peroxisome proliferator-activated receptor-(PPAR-) agonist that also counteracts SARS-CoV-2 entry and its replication. Our work highlights for the first time the antiviral potency of PEA against SARS-CoV-2, exerting its activity by two different mechanisms. First, its binding to the SARS-CoV-2 S protein causes a drop in viral infection of ~70%. We show that this activity is specific for SARS-CoV-2, as it does not prevent infection by VSV or HSV-2, other enveloped viruses that use different glycoproteins and entry receptors to mediate their entry. Second, we show that in infected Huh-7 cells, treatment with PEA dismantles lipid droplets, preventing the usage of these vesicular bodies by SARS-CoV-2 as a source of energy and protection against innate cellular defenses. This is not surprising since PEA activates PPAR-, a transcription factor that, once activated, generates a cascade of events that leads to the disruption of fatty acid droplets, thereby bringing about lipid droplet degradation through-oxidation. In conclusion, the present work demonstrates a novel mechanism of action for PEA as a direct and indirect antiviral agent against SARS-CoV-2. This evidence reinforces the notion that treatment with this compound might significantly impact the course of COVID-19. Indeed, considering that the protective effects of PEA in COVID-19 are the current objectives of two clinical trials (NCT04619706 and NCT04568876) and given the relative lack of toxicity of PEA in humans, further preclinical and clinical tests will be needed to fully consider PEA as a promising adjuvant therapy in the current COVID-19 pandemic or against emerging RNA viruses that share the same route of replication as coronaviruses.

It is currently used for the treatment of different types of neuropathic pain, such as fibromyalgia, osteoarthritis, carpal tunnel syndrome, and many other conditions. The properties of PEA, especially of its micronized or ultra-micronized forms maximizing bioavailability and efficacy, have sparked a series of innovative research to evaluate its possible application as therapeutic agent for neurodegenerative diseases. Neurodegenerative diseases are widespread throughout the world, and although they are numerous and different, they share common patterns of conditions that result from progressive damage to the brain areas involved in mobility, muscle coordination and strength, mood, and cognition. The present review is aimed at illustrating in vitro and in vivo research, as well as human studies, using PEA treatment, alone or in combination with other compounds, in the presence of neurodegeneration. Abstract:  Intracerebral hemorrhage is a type of acute cerebrovascular disease that remains one of the main causes of death and disability. After the onset of ICH, different types of severe pathophysiological changes can cause great damage to brain tissue, including neuroinflammation. Our study demonstrated the effect of PEA on modulating microglia phenotype and neuroinflammation, as well as the possible underlying mechanisms after ICH for the first time. The phenotypic transformation of microglia and simulation of neuroinflammation after ICH in vitro was induced by hemoglobin on BV2 cells. Additionally, the experiment in vivo model was induced by collagenase injection in mice. The role of PEA on hematoma clearance was also discussed. Western blot, ELISA and immunofluorescence staining were used to determine the phenotypic polarization of microglia and neuroinflammation. In order to evaluate the role of PPAR-in the anti-inflammatory effect of PEA after ICH, the PPAR-antagonist GW6471 was utilized. Behavior tests examined the effect of PEA on improving neuronal function. Our results showed that PEA can ameliorate neuroinflammation by inhibiting upregulation of NF-B, IL-1and TNF-, both in vivo and in vitro. Additionally, PEA can improve motor function in ICH mice and promotes hematoma clearance. At the same time, PEA can increase the levels of PPAR-in the nucleus. Hence, PPAR-antagonists can reverse the protective effects of PEA on neuroinflammation. These results suggest that PEA is involved in microglia polarization, attenuating the activation of neuroinflammation, as well as improving motor function after ICH. This, at least in part, may contribute to the involvement of PPAR-modulation of NF-B.

In some cases of COVID-19, patients can be characterized by clinical sequelae with mild-to-moderate symptoms that persist long after the resolution of the acute infection, known as long-COVID, potentially affecting their quality of life. The main symptoms of long-COVID include persistent dyspnea, fatigue and weakness (that are typically out of proportion, to the degree of ongoing lung damage and gas exchange impairment), persistence of anosmia and dysgeusia, neuropsychiatric symptoms, and cognitive dysfunctions (such as brain fog or memory lapses). The appropriate management and prevention of potential long-COVID sequelae is still lacking. It is also believed that long-term symptoms of COVID-19 are related to an immunity over-response, namely a cytokine storm, involving the release of pro-inflammatory interleukins, monocyte chemoattractant proteins, and tissue necrosis factors. Palmitoylethanolamide (PEA) shows affinity for vanilloid receptor 1 and for cannabinoid-like G protein-coupled receptors, enhancing anandamide activity by means of an entourage effect. Due to its anti-inflammatory properties, PEA has been recently used as an early add-on therapy for respiratory problems in patients with COVID-19. It is believed that PEA mitigates the cytokine storm modulating cell-mediated immunity, as well as counteracts pain and oxidative stress. In this article, we theorize that PEA could be a potentially effective nutraceutical to treat long-COVID, with regard to fatigue and myalgia, where a mythocondrial dysfunction is hypothesizable.

Abstract:  Ultramicronized palmitoylethanolamide (um-PEA), a compound with antioxidant, anti-inflammatory and neuroprotective properties, appears to be a potential adjuvant treatment for early stages of Coronavirus disease 2019 (COVID-19). In our study, we enrolled 90 patients with confirmed diagnosis of COVID-19 that were randomized into two groups, homogeneous for age, gender and BMI. The first group received oral supplementation based on um-PEA at a dose of 1800 mg/day for a total of 28 days; the second group was the control group (R.S. 73.20). At baseline (T0) and after 28 days of um-PEA treatment (T1), we monitored: routine laboratory parameters, inflammatory and oxidative stress (OS) biomarkers, lymphocytes subpopulation and COVID-19 serological response. At T1, the um-PEA-treated group presented a significant reduction in inflammation compared to the control group (CRP= 0.007; IL-6= 0.0001; neutrophils to lymphocytes ratio= 0.044). At T1, the controls showed a significant increase in OS compared to the treated group (FORT= 0.05). At T1, the um-PEA group exhibited a significant decrease in D-dimer levels (= 0.0001) and higher levels of IgG against SARS-CoV-2 (= 0.0001) compared to the controls. Our data demonstrated, in a randomized clinical trial, the beneficial effects of um-PEA in both asymptomatic and mild-symptomatic patients related to reductions in inflammatory state, OS and coagulative cascade alterations.

The pathogenesis of DIC is complex, with cross-talk between the coagulant and inflammatory pathways. The objective of this study is to investigate the anti-inflammatory action of ultramicronized palmitoylethanolamide (um-PEA) in a lipopolysaccharide (LPS)-induced DIC model in rats. Experimental DIC was induced by continual infusion of LPS (30 mg/kg) for 4 h through the tail vein. Um-PEA (30 mg/kg) was given orally 30 min before and 1 h after the start of intravenous infusion of LPS. Results showed that um-PEA reduced alteration of coagulation markers, as well as proinflammatory cytokine release in plasma and lung samples, induced by LPS infusion. Furthermore, um-PEA also has the effect of preventing the formation of fibrin deposition and lung damage. Moreover, um-PEA was able to reduce the number of mast cells (MCs) and the release of its serine proteases, which are also necessary for SARS-CoV-2 infection. These results suggest that um-PEA could be considered as a potential therapeutic approach in the management of DIC and in clinical implications associated to coagulopathy and lung dysfunction, such as COVID-19.

A comicronized formulation of ultramicronized PEA (um-PEA) and polydatin (Pol) PEA/Pol, a biological precursor of resveratrol with antioxidant activity, could have protective effects on oxidative stress produced by inflammatory processes. We evaluated the effects of a comicronized PEA/Pol 10 mg/kg (9 mg of um-PEA+1 mg of polydatin) in a model of Dinitrobenzene sulfonic acid (DNBS)-induced colitis. Ulcerative colitis was induced in mice by intrarectally injection of DNBS (4 mg in 100L of 50% ethanol per mouse). Macroscopic and histologic colon alterations and marked clinical signs were observed four days after DNBS and elevated cytokine production. The myeloperoxidase (MPO) activity assessed for neutrophil infiltration was associated with ICAM-1 and P-selectin adhesion controls in colons. Oxidative stress was detected with increased poly ADP-ribose polymerase (PARP) and nitrotyrosine positive staining and malondialdehyde (MDA) levels in inflamed colons. Macroscopic and histologic alterations minimized by oral PEA/Pol, as well as neutrophil infiltration, inflammatory cytokine release, MDA, nitrotyrosine, PARP and ICAM-1, and P-selectin expressions. The mechanism of action of PEA/Pol could be related to the sirtuin 1/nuclear factor erythroid 2-related factor 2 (SIRT-1/Nrf2) pathway and nuclear factor (NF)-B. PEA/Pol administration inhibited NF-B and increased SIRT-1/Nrf2 expressions. Our results show that PEA/Pol is capable of decreasing inflammatory bowel disease (IBD) DNBS-induced in mice.

In the present study, we tested different associations of um-PEA, LAC and non-micronized PEA (non-m-PEA) in a rat model of carrageenan (CAR)-induced paw edema. Intraplantar injection of CAR into the hind paw of animals caused edema, thermal hyperalgesia, accumulation of infiltrating inflammatory cells and augmented myeloperoxidase (MPO) activity. All these parameters were decreased in a significantly manner by oral administration of a compound constituted by a mixture of um-PEA and LAC in relation 1:1 (5 mg/kg), but not with the association of single compounds administered one after the other. These findings showed the superior anti-inflammatory and anti-nociceptive action displayed by oral administration of um-PEA and LAC versus LAC plus, separate but consecutive, um-PEA in the rat paw CAR model of inflammatory pain.

Bacopa monnieri was observed to inhibit the Tau aggregation in vitro. The cells exposed to Bacopa monnieri were also observed to have a low level of ROS and caspase-3 activity. The immunoblot and immunofluorescence analysis showed that Bacopa monnieri acts as an antioxidant and restored the Nrf2 levels in Neuro2a cells. Bacopa monnieri treatment to Neuro2a cells was observed to reduce the phospho-Tau load in formaldehyde-stressed cells. Furthermore, the treatment of Bacopa monnieri reduced the phosphorylation of GSK-3in formaldehyde-stressed cells. Ran and NUP358 are the key proteins involved in nuclear transport. It was observed that formaldehyde treatment impaired the nuclear transport by missorting the NUP358 arrangement in Neuro2a cells. On the contrary, Bacopa monnieri treatment restored the NUP358 arrangement in cells. The overall results of the present study suggested that Bacopa monnieri could be considered a potent herb against Tau phosphorylation and Tau aggregation, which projects it as a promising formulation for Alzheimer's disease.

However, the effects of CBD, its mechanism of action, and modulation of downstream signaling pathways in urothelial cells, the main effector cells in IC/BPS, have not been fully elucidated yet. Here, we investigated the effect of CBD against inflammation and oxidative stress in an in vitro model of IC/BPS comprised of TNF-stimulated human urothelial cells SV-HUC1. Our results show that CBD treatment of urothelial cells significantly decreased TNF-upregulated mRNA and protein expression of IL1, IL8, CXCL1, and CXCL10, as well as attenuated NFB phosphorylation. In addition, CBD treatment also diminished TNF-driven cellular reactive oxygen species generation (ROS), by increasing the expression of the redox-sensitive transcription factor Nrf2, the antioxidant enzymes superoxide dismutase 1 and 2, and hem oxygenase 1. CBD-mediated effects in urothelial cells may occur by the activation of the PPARreceptor since inhibition of PPARresulted in significantly diminished anti-inflammatory and antioxidant effects of CBD. Our observations provide new insights into the therapeutic potential of CBD through modulation of PPAR/Nrf2/NFB signaling pathways, which could be further exploited in the treatment of IC/BPS.

The objective of this study was to examine the anti-inflammatory effect of cannabidiol in combination with M. citrifolia and compare its effects with those of cannabidiol alone.MATERIALS AND METHODS: RAW264 cells stimulated with lipopolysaccharide (200 ng/ml) were treated with cannabidiol (0-10M), M. citrifolia seed extract (0-100g/ml), or a combination of both for 8 or 24 h. Following the treatments, nitric oxide production in the activated RAW264 cells and the expression of inducible nitric oxide synthase were assessed.RESULTS: Our results showed that combination of cannabidiol (2.5M) and M. citrifolia seed extract (100g/ml) exhibited more efficient inhibition of nitric oxide production than cannabidiol treatment alone in lipopolysaccharide-stimulated RAW264 cells. As questions about the safety of the experimental gene transfer shots mounted, another campaign slogan was concocted: Dont do your own research. At the same time, Trust the science was trending. What that meant was that you were supposed to trust that what you were told WAS the science. Actually looking at published science, that made you a dangerous moron.

Common fennel (Foeniculum vulgare) is a perennial plant in the carrot family that is grown for the edible leaves, shoots, seeds and bulbs. It's likely the plant was first cultivated in Israel,1 and is believed to have been part of Jewish meals since biblical times. If the seed heads are collected before they shatter, it may also help prevent the spread of the plant. In this study, we aimed to determine whether MOOs can improve the symptoms of depression in individuals with hypertension.METHODS: Dahl salt-sensitive rats fed with a high-salt diet were stimulated by chronic unpredictable mild stress to mimic hypertension with depression. Primary astrocytes and neurons were isolated from these rats. Astrocytes underwent LPS stimulation to simulate the inflammatory astrocytes during depression. MOOs were administrated at 0.1 mg/g/day in vivo and 1.25, 2.5, and 5 mg/mL in vitro. Mitophagy was inhibited using 5 mM 3-methyladenine (3-MA). Astrocyte-mediated neurotoxicity was detected by co-culturing astrocytes and neurons.RESULTS: MOOs decreased systolic pressure, diastolic pressure, and mean arterial pressure, thereby improving depression-like behavior, including behavioral despair, lack of enthusiasm, and loss of pleasure during hypertension with depression. Furthermore, MOOs inhibited inflammation, astrocytic dysfunction, and mitochondrial damage in the brain. Then, MOOs promoted autophagosome and lysosome enriched in mitochondria in LPS-stimulated astrocytes. MOOs suppressed mitochondrial damage and the release of tumor necrosis factor-(TNF-), interleukin (IL)-6, and IL-1in astrocytes undergoing LPS stimulation. Importantly, MOOs rescued the impaired neurons co-cultured with astrocytes. The effects of MOOs on LPS-stimulated astrocytes were reversed by 3-MA. Finally, MOOs upregulated LPS-downregulated Mfn2 expression in astrocytes. Mfn2 inhibition partly reversed the effects of MOOs on hypertension with depression. Intriguingly, Mfn2 suppression activated PI3K/Akt/mTOR pathway during MOOs treatment.CONCLUSIONS: Astrocytes develop neuroinflammation in response to mitochondrial damage during hypertension with depression. Abstract:  Heart failure (HF) is a cardiovascular disease with an extremely high mortality rate. However,How (MO) has not been studied for cardiovascular purposes at this time, the aim of this study was to find new mechanism for the MO of treatment of HF through a bioinformatics and experimental validation. The present study also aimed to establish a link between the basic and clinical applications of this medicinal herb. MO compounds and targets were obtained by traditional Chinese medicine systems pharmacology (TCMSP) and Pubchem. Subsequently, HF targets were acquired from DisGeNET and the interactions of all the targets and other human proteins were obtained via String so as to establish a component-target interaction network by Cytoscape 3.7.2. All the targets of clusters were inserted into Database for Annotation, Visualization and Integrated Discovery (DAVID) to perform GO (gene ontology) enrichment analysis. Molecular docking was adopted to predict the targets of MO relevant to the treatment of HF and to further explore the associated pharmacological mechanisms. Subsequently, a series ofexperiments, including histopathological staining, immunohistochemical and immunofluorescence analyses were conducted for further verification. Moreover, western blot analysis andexperiments were performed. The results indicated that MO alleviated apoptosis, regulated cholesterol metabolism and transport function, and reduced inflammation, which resulted in the successful treatment of HF. Beta-sitosterol, Asperuloside tetraacetate and americanin A were the key bioactive components of MO. ALB, AKT1, INS, STAT3, IL-6, TNF, CCND1, CTNNB1, CAT, and TP53 were the core potential targets, which were significantly associated with multiple pathways, namely the FoxO signaling pathway, the AMPK signaling pathway, and the HIF-1 signaling pathway.experiments validated that MO may protect against heart failure or treat this disease by increasing the levels of autophagy via the FoxO3 signaling pathway in rats. The present study suggested that a combination of network pharmacology prediction with experimental validation may offer a useful tool to characterize the molecular mechanism of action of the traditional Chinese medicine (TCM) MO in the treatment of HF.

Skin pathological analysis and ELISA assay were used to detect the effect of MOE on the inflammation of AD model mouse skin and the expression changes of inflammatory factors, and further functional verification was performed in TNF-/IFN--induced HaCaT cells.RESULTS: Our in vivo experiments confirmed that MOE remarkably reduced DNCB-induced AD lesions and symptoms, such as epidermal and dermal thickness and mast cell infiltration and inflammatory cytokines secretion in the mice models. In addition, the underlying mechanisms by which MOE ameliorated AD had been uncovered, and we verified that MOE inhibited MALAT1 expression in AD, resulting in attenuated expression of C-C chemokine receptor type 7 (CCR7) regulated by MALAT1-sponge miR-590-5p in a competing endogenous RNA (ceRNA) mechanisms-dependent manner, thereby inhibiting TNF-/IFN--induced cellular proliferation and inflammation.

All the isolates were evaluated for their inhibitory effect on nitric oxide (NO) production induced by lipopolysaccharide (LPS) in murine RAW264.7 macrophage cells, and the enantiomers 1a and 3b exhibited moderate activity with ICvalues of 41.9 and 26.2 M. Meanwhile, compound 3b also dose-dependently inhibited the secretion of two pro-inflammatory cytokines TNF-and IL-6 in the same cell model.

In this study, we investigated the preventive effect ofleaf (NNL) ethanolic extract on muscle atrophy induced by dexamethasone (DEX), a synthetic glucocorticoid, in mice and its underlying mechanisms. The administration of NNL extract increased weight, cross-sectional area, and grip strength of quadriceps (QD) and gastrocnemius (GA) muscles in DEX-induced muscle atrophy in mice. The NNL extract administration decreased the expression of muscle atrophic factors, such as muscle RING-finger protein-1 and atrogin-1, and autophagy factors, such as Beclin-1, microtubule-associated protein 1A/1B-light chain 3 (LC3-I/II), and sequestosome 1 (p62/SQSTM1) in DEX-injected mice. DEX injection increased the protein expression levels of NOD-like receptor pyrin domain-containing protein 3 (NLRP3), cleaved-caspase-1, interleukin-1beta (IL-1), and cleaved-gasdermin D (GSDMD), which were significantly reduced by NNL extract administration (500 mg/kg/day). In vitro studies using C2C12 myotubes also revealed that NNL extract treatment inhibited the DEX-induced increase in autophagy factors, pyroptosis-related factors, and NF-B. Overall, the NNL extract prevented DEX-induced muscle atrophy by downregulating the ubiquitin-proteasome system, autophagy pathway, and GSDMD-mediated pyroptosis pathway, which are involved in muscle degradation.

Lotus root methanol extract were identified by UPLC-QTOF-MS/MS based metabolomics analysis. Then 109 active ingredients with achievable oral doses and drug-like properties were explored using the TCMSP platform. SwissTargetPrediction Database to predict lotus root treatment targets for acute alcoholismSTRING database (https://www.string-db.org/) was used to construct protein-protein interaction network graphs. Gene ontology (GO) functional, Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of genes common to lotus root and alcoholism by Metascap database. Molecular docking simulations were performed using AutoDock 1.5.6 software. Animal experiments verified the relieving effect of lotus root extract on acute alcoholism after intervention.RESULTS: Results indicated the methanol extract of lotus root showed the highest activation rate of ethanol dehydrogenase(18.87%). The 433 compounds of lotus root methanol extract were identified by UPLC-QTOF-MS/MS based metabolomics analysis. Bioinformatics analysis indicate that there were 224 intersectioning targets between lotus root extract and acute alcoholism. KEGG enrichment analysised shows that lotus root extract may play a role in treating acute alcoholism by intervening with the neuroactive ligand-receptor interaction pathway. The protein-protein interaction network (PPI) analysis found that HSP90AA1, MAPK1 and STAT3 played a key role in lotus root extract-modulated PPI networks. Molecular docking showed that (7R, 8S)-dihydrodihydrodipine cypressol had the best binding ability with MAPK1. Experiments in mice indicate that lotus root extract improved the activity of liver alcohol dehydrogenase (ADH), acetaldehyde dehydrogenase (ALDH), catalase (CAT), superoxide dismutase (SOD) and glutathione peroxidase (GSH-PX), increase glutathione (GSH) and reduce malondialdehyde (MDA) levels, decrease glutamate transaminase (AST), alanine transaminase (ALT) and alkaline phosphatase (AKP) in the serum of mice with acute alcoholism, and accelerate the metabolic rate of alcohol after drinking. This week, we enter the month of June and have a Sagiartius Full Moon on June 3rd. The cards guide us and call our best selevs forward as we journey through the days ahead.

Our first card this week is the Queen of Wands. She is the fire inside all of us. She is the energy that makes us loving and loyal friends and worthy adversaries.

The Queen speaks to the passionate energy that bubbles up when we see injustices in the world. And the warrior in us that has been through the very worst of days and made it out stronger.

This makes her tough and opinionated but lacking when it comes to communicating in a diplomatic way. She can be overpowering and turn people off with her intensity.

As we journey through Gemini Season, it is even more important we are aware of how we communicate, as it can make it even more challenging. We should use the lessons of the Queen of Wands to practice compassionate expression.

This week we are called to connect to our fire within while learning to be aware of how this fire might impact others. Some questions we might ask are:

How can we be both strong and warm at the same time? How can we share our passion without alienating others? How can we have compassion for those we disagree with while still holding them accountable?

Let the fire push you to create change through understanding and grace. The kindness in your communication and actions will make them so much more impactful.

Our next card this week is The Moon. The Moon is number eighteen in the Major Arcana and depicts a mysterious night-time scene. The card here shows Anubis, the jackal god of the Egyptians. Below is a scarab beetle holding a glowing moon.

The scarab is is a symbol of renewal and immortality, portaying the Moons association with natural cycles and rhythms. We are called to tap into the cycles and rhythms of our lives and to allow ourselves to go with the flow.

When we resist the natural ups and downs, we find ourselves exhausted and out of sync. But when we accept that everything has both peaks and valleys, we are able to feel more grounded and at peace.

We know that after winter there is always spring. And that all New Moons will lead into a Full Moon and back again. It is the same with our lives. There are times of plenty and times with less. The more comfortable we are with the cyclical nature of life the more we can find the good in each moment.

The Moon card indicates a voyage to the centre of ourselves. When we journey inside, we dont find flaws or weaknesses, but an unshakeable core that is steady through all the phases of life.

Abstract:  High blood concentrations of nonesterified fatty acids (NEFAs) provoke various metabolic disorders and are associated with mammary tissue injury and decreased milk production in dairy cows. Nuciferine, an alkaloid found inleaves, has great potential for correcting lipid metabolism derangements and lipotoxicity. In this study, we evaluated the lipotoxicity induced by excessive NEFA in bovine mammary epithelial cells (bMECs) and investigated whether nuciferine alleviates NEFA-induced lipotoxicity and the underlying molecular mechanisms. We found that excessive NEFA (1.2 and 2.4 mM) induced lipid accumulation, apoptosis, and migration ability impairment in bMECs, whereas nuciferine could ameliorate these disarrangements, as indicated by decreasing triglyceride content, protein abundance of SREBP-1c, cytoplasmic cytochrome, and cleaved caspase-3 and increasing protein abundance of PPARand migration ability. Moreover, nuciferine could reverse NEFA-induced LKB1/AMPK signaling inhibition, and the protective effect of nuciferine on lipotoxicity caused by NEFA was abrogated by AMPK inhibitor dorsomorphin. Furthermore, transfection with LKB1 siRNA (si-LKB1) largely abolished the activation effect of nuciferine on AMPK. Overall, nuciferine can protect bMECs from excessive NEFA-induced lipid accumulation, apoptosis, and impaired migration by activating LKB1/AMPK signaling pathway.

The prevention and management of myocardial ischemia/reperfusion (I/R) injury is an essential part of coronary heart disease surgery and is becoming a major clinical problem in the treatment of ischemic heart disease. Nuciferine has potent anti-inflammatory and antioxidative stress effects, but its role in myocardial ischemia-reperfusion (I/R) is unclear. In this study, we found that nuciferine could reduce the myocardial infarct size in a mouse myocardial ischemia-reperfusion model and improve cardiac function. Furthermore, nuciferine could effectively inhibit hypoxia and reoxygenation (H/R) stimulated apoptosis of primary mouse cardiomyocytes. In addition, nuciferine significantly reduced the level of oxidative stress. The peroxisome proliferator-activated receptor gamma (PPAR-) inhibitor GW9662 could reverse the protective effect of nuciferine on cardiomyocytes. These results indicate that nuciferine can inhibit the apoptosis of cardiomyocytes by upregulating PPAR-and reducing the I/R-induced myocardial injury in mice.

Nuciferine alleviates intestinal inflammation. "IndyWatch Feed Allcommunity"

Abstract:  Nuciferine (NCF) is an aporphine alkaloid and a principal bioactive constituent in the lotus plant. Herewith, we investigated the potential anti-inflammatory effect and underlying mechanisms of NCF employing dextran sulfate sodium (DSS)-induced ulcerative colitis in mice, a predominant intestinal inflammatory disease, and mouse RAW 264.7 cells in vitro. Lipopolysaccharide (LPS) was used to generate an inflammatory response in the RAW 264.7 cells. The disease activity index (DAI), colon morphology, colonoscopy, and colon histopathology were performed to assess experimental colitis. The biochemical assays, enzyme-linked immunosorbent assay (ELISA), and immunoblot analysis were performed to understand the underlying mechanisms. In RAW 264.7 cells, NCF pretreatment significantly decreased the expression of inducible nitric oxide synthase (iNOS), the expression and release of pro-inflammatory cytokines including interleukin (IL)-1, IL-18, and tumor necrosis factor-(TNF-) and interfered with the activation of mitogen-activated protein kinase (MAPK), nuclear factor-B (NF-B), and NOD-like family pyrin domain containing 3 (NLRP3) signaling pathways. The oral treatment of NCF substantially alleviated the DSS-induced DAI, increased colon length, and restored colon morphology and histology. Compared to the DSS-induced mice, the proteins involved in the activation of MAPK/NF-B/NLRP3 pathways and the cytokines were markedly decreased in the NCF-treated mice. Moreover, the tight junction architecture of the colon was well-maintained in NCF treatment groups by regulating the expression of claudin-1 and zonula occludens-1 (ZO-1) proteins. All these findings suggest that NCF can be a promising molecule to modulate ulcerative colitis.

Massive Sunspot Four Times the Size of Earth Is Now Visible Without a Telescope "IndyWatch Feed Allcommunity"

The size of one of the suns sunspots has increased to the point that it can be seen from Earth without the need of magnification; nevertheless, eye protection is still clearly recommended while viewing it.

It is no longer necessary to use a telescope in order to see sunspot AR3310 from Earth since its size has increased to the point where it is now visible with the naked eye. An image of the sunspot, with a little Earth superimposed on it, was released on the internet by a South Korean astronomer by the name of Bum-Suk Yeom.

Naked-Eye Sunspot using Solar Glasses (or Solar Filter), Yeom said in an Instagram post sharing the image. A giant sunspot, visible to the naked eye, is crossing the suns disk. A [sunspot] can be seen to the left of the center of the solar disk.

Sunspots are patches of darkness that appear on the surface of the sun and are created by concentrations of magnetic fields that are exceptionally strong and twisted. If these fields are strong enough, they prevent part of the heat from the suns core from reaching its atmosphere, which results in the surface of the sun being colder and seeming darker than normal.

Sometimes sunspots are big enough that they may be seen with the naked eye, however you should still use eye protection since the suns glare can be rather intense.
