Large Scale Thread Portraits Are Masterfully Embroidered on Tulle "IndyWatch Feed Allcommunity"

Large Scale Thread Portraits Are Masterfully Embroidered on Tulle

Embroidery Art by Katerina Marchenko

We are accustomed to seeing embroidery made on a small scale, but artist Katerina Marchenko takes this art form and makes it lifesize. Using colorful strands of yarn, she renders massive human portraits on delicate tulle and showcases the endless possibilities this medium has to offer.

Marchenko shows how she approaches each piece via her social media. In these videos, we see her weave long and wide pieces of thread through the tulle using her hands (san needle). She begins by outlining the facial features like the eyes, nose, and mouth with black. Then, she adds various hues to create planes of color, filling the portrait with a sense of dimension. However, rather than trying to disguise her method, she highlights the thread medium by adding stylistic touches to these faces in the form of long loose threads which cascade down the face or radiate from the head.

These large-scale portraits possess similar qualities to expressive paintings. The way in which Marchenko manipulates the array of different colored threads mimics brushstrokes on a canvas. And by alternating between light and dark shades, she even models these human figures.

You can purchase original embroidery creations via Marchenko's Etsy store and keep up to date with her latest creations by following the artist on Instagram.

Fascinating Animation Compares the Size of Extinct Animas With Their Descendants "IndyWatch Feed Allcommunity"

Fascinating Animation Compares the Size of Extinct Animas With Their Descendants

When we think of the size of extinct animals, our minds might go to the large T-Rex in Jurassic Park. As frightening as that looks, there are other creatures that, while long gone, would've been quite scary to encounter. YouTube channel WTD has created a digital animation showcasing the sizes of extinct animals alongside their extant descendants. Let's just say that some daunting creatures of today had much more intimidating relatives in the long-since past.

The video begins presenting the nuralagus, a yard-length relative of the rabbit. Things, however, quickly escalate from there. For example, while modern beavers are about 17 inches tall, their ancestor, the castoroides, was over three feet tall and eight feet long. The same goes for the Emperor penguin. At four feet tall, they are endearing, but the Anthropornis Nordenskjoeldi was six feet highso basically as tall as a professional basketball player.

Some threatening creatures currently in the world make an appearance alongside their ancestors. The 32-foot reticulated python is joined by the 45-foot titanoboa, while the saltwater crocodile, at about 10 feet long, is four times smaller than the Rhamphosuchus, which was 39 feet long.

The animation also features some creatures that, while almost similar in size to their descendants, had some very distinctive features. This includes the megaloceros' large antlers, which rival the size of a moose, or the arctotherium, a bear almost similar in height to the polar bear but with dark fur and native to Central and South America.

WTD, which is dedicated to collecting and making data comparison of all things in 3D, has also produced some other insightful animations comparing animals present and past. To stay up to date with its videos, you can subscribe to its YouTube channel.

I prefer leather gloves. Haven't found one that is reasonably priced, rugged, comfortable, not too thick and has a loop or hole to tie on to a carabiner on my pack. Suggestions?

My current gloves are cheap, rugged, fairly comfortable, but not attachment point. This is maybe my fourth or fifth pair? I usually will use them to death then replace. Tried a pair that was partly leather, but did not like it as much. The fingertip dexterity wasn't that good on that pair.

Tron (TRX) Sees 10% Price Surge Following Bittorrent Bridge Launch "IndyWatch Feed Allcommunity"

Tron (TRX), the native cryptocurrency of the Tron blockchain, witnessed a substantial increase over the weekend. The token spiked by 10% yesterday, buoyed by recent developments within its ecosystem.  However, at the time of writing, it has experienced a slight correction, but all signs show that this could be a bullish signal.

Tron Spike Due To Bittorent Bridge Launch

The reason behind this latest surge can be attributed to the recent launch of the BitTorrent Bridge platform, which now allows users to seamlessly transfer their TRX tokens between the Tron blockchain and Ethereum. Justin Sun, the founder of Tron, confirmed this development on the morning of June 2.

The BitTorrent Bridge serves as a decentralized system that facilitates token interoperability across different blockchain networks. Developed by the Tron organization under the leadership of Justin Sun, this platform strengthens the collaborative efforts between BitTorrent and Tron. A notable instance of their collaboration is the BitTorrent Live streaming service, which was launched in 2019 and operates in conjunction with Trons system.

Related Reading: Quant Hype Spikes As Price Surges 15% Is This A Top Signal?

The integration between TRON and Ethereum through the BitTorrent Bridge opens up new possibilities for TRX users while enhancing the interoperability between these prominent blockchains. This breakthrough expands Trons presence within the Ethereum ecosystem and has the potential to drive greater adoption of TRX among Ethereum users.

It is important to note that TRX initially existed as an ERC-20 token on the Ethereum network before being migrated to the TRON main network in mid-2018. Since then, Tron has grown into one of the biggest blockchains and currently processes 40% of USDT transactions.

Related Reading: XRP Climbs 12% As Crypto Celebrates 11-Year Milestone

Tron Price Analysis

Analyzing Trons current price performance, the coin is currently trading at $0.0805. Moreover, Tron has the potential to maintain its position above the moving averages. If the resistance level of $0.085 is breached, it could serve as a catalyst for a bullish trend, potentially leading to further price gains.

The market sentiment appears optimistic as buyers continue to demonstrate a strong commitment. Based on the daily chart analysis, another upward movement in the market price is possible. Therefore, if Tron surpasses the channels upper boundary, it could further reinforce the market and reach resistance levels of $0.095, $0.100, and $0.105, respectively.

Three Reasons Behind Bitcoin's 2023 Resurgence "IndyWatch Feed Allcommunity"

This is an opinion editorial by Chen Fang, the COO of BitGO, a Bitcoin-focused regulated custody and financial services firm.

A challenging 2022 prompted Bitcoin skeptics to gleefully say I told you so and declare that Bitcoin was dead and buried. But, much to their chagrin, their victory lap ended up being premature. Bitcoin has come roaring back in 2023, ending these premature celebrations and taking back some of its 2022 losses with a massive year-to-date gain of over 60% as of this writing so far in 2023.

Here are three reasons behind Bitcoins 2023 resurgence:

One: Banking Sector Turmoil And A Timely Reminder Of Bitcoins Value

In March 2023, turmoil from the banking sector in the wake of Silicon Valley Banks collapse rocked the market with the types of issues that it hadnt experienced since the Great Recession. This was no fly-by-night operation; at the time, Silicon Valley Bank (SVB) was the sixteenth-largest bank in the United States and the bank of choice for many venture capitalists and startups.

This high-profile implosion and the lack of proper risk management in place at SVB led many individuals to question how safe their money was. Ultimately, the Federal Reserve stepped in to make the banks depositors whole, but the event still called the health of the banking system into question.

While not everyone is rushing to take their money out of banks, many more people are now at least aware of the risks of a single point of failure and are moving to diversify their assets by splitting them between multiple banks or even diversifying into alternatives like bitcoin and other cryptocurrencies for the first time.

The appeal of a fully decentralized asset like bitcoin is that there is no CEO or management team in charge that can endanger the solvency of the Bitcoin network by making a bad decision or poor judgment call. Bitcoin users dont have to trust a company or management team to use Bitcoin; they can instead verify the open-source code that governs the Bitcoin network. Anyone can view any transaction ever made on the Bitcoin blockchain, giving the network unmatched transparency.

The price of bitcoin surged in March in the wake of the crisis, and in late April, renewed troubles at First Republic Bank, which reported that it...

Cult Conspiracy: US Charges Two in Plot Against Chinese Religious Group "IndyWatch Feed Allcommunity"

Two Chinese nationals allegedly acting as agents for the Chinese government were apprehended in the United States following an undercover operation and were charged with trying to damage and topple an anti-communist Chinese religious group.

Authorities in Los Angeles arrested 70-year-old John Chen and 43-year-old Lin Feng on May 26th, where they were indicted for planning to revoke the tax-exempt status of the New York-based Falun Gong spiritual movement in an attempt to further undermine the exiled group.

The two Chinese nationals were both born in China but lived in Los Angeles at the time of their arrest. Chen is a US citizen, while Feng was described as a lawful permanent resident. Theyre facing charges of being unregistered agents of a foreign government, practicing money laundering, and bribing public officials.

Chen and Feng were caught after they initially urged the Internal Revenue Service (IRS) to revoke Falun Gongs non-profit tax status. Chen also described the organization as a gigantic mega cult in his whistleblower complaint that he filed to the tax agency last February, echoing the language used by the Chinese Communist Party (CCP) against the group.

After filing their complaint, an undercover police officer posed as an official for the IRS. According to prosecutors handling their case, Chen met with the officer at a restaurant north of New York City on May 14th.

Chen approached the officer to ensure that the agency would act on their complaint while communicating with h...

OP here is from a strict Muslim background, though he claims to be not that religious himself. He mentions that his girlfriend is American and not the modest kind, which has caused a few awkward bumps in what he describes as an otherwise smooth road.

So me (26M) and my Girlfriend (22F) have been dating for around 2 years now. Weve had our ups and downs but overall we have a great relationship, and I definitely see a future together! We really love and respect each other and connect well! But the thing is we come from different backgrounds. I come from a Muslim Background (Im not that religious tho) and she is White. However, we have still made it work and our similarities are much more than our differences.

Now my gf does like wearing a bit short clothing when out and about, which used to bother me at first and made me uncomfortable and we did have a few minor fights over it, but eventually I came to terms with it.

Recently they had a bump that he describes as a bit embarrassing, and it happened at his familys pool party.

He spoke with her ahead of time about making a modest bathing suit choice, and so was shocked to see her arrive in a cheeky bikini. OP was so mortified he couldnt look at or speak to his family!

However, recently an embarrassing situation happened. There was a pool party at my cousins house, and my parents were there too. I had recently introduced my gf to my parents as well, so this was a good time for to meet family and relatives. I specifically told her to please pack somet...

Punk Boy Scout Helps Elderly Woman Across Open Pit "IndyWatch Feed Allcommunity"

OCEANVIEW, Ill. Local sixth-grade do-gooder Richie Amweather reportedly risked his own well-being at last nights hardcore show in order to help a geriatric woman across the pit, heartwarmed sources confirmed.

Speaking as a dedicated member of Troop 138, I thought not only was it the right thing to do, but it would count a long way toward being awarded the Pit Etiquette merit badge Ive been going for the past year. So, I offered my arm the instant I saw her narrowly avoid getting crowd punched, said 11-year-old Amweather. Sure, I got elbowed in the face a couple dozen times, and I got five or six tall boys spilled on my dry-clean only uniform, but it was all worth it to know that sweet old lady was safe from harms way. And the pat on the head she gave me after? Icing on the cake, yes sir!

The elderly woman in question, Ms. Lilac McMurtree, was reportedly more than appreciative for the scouts assistance.

Oh, that sweet young man gives me hope for the future. I wandered in after my trip to the greengrocer, intrigued by the look of the young combo on stage yelling fuck this and fuck that. I had never seen an amplifier colored orange, and wanted to get a closer look, said the 83-year-old retired sheet music store employee. With my hearing aid out of battery, I relied on little Richies guidance to get me up past the fine people kneeing each other in the back. I tried to give him a shiny nickel later to get himself an EP at the merch table, but he refused, saying doing good was its own reward.

Naming the program after his favorite krautrock band, Scout Master Wilson Hauer elaborated further on the Neu! Scout ethos.

That Amweather boy is turning into a fine young punk. Oughta complete Neu! Scouts at the top of his troop, said Hauer, while casually fashioning a microphone out of a stolen payphone receiver. Especially if he sells enough of my bands demo tapes outside the mini-mart come fundraising time. I tell you, these kids may not be old enough to get into a lot of the venues our trips take us to, but the xs on their hands are no match for the ps and qs they learn to always mind.

At press time, Amweather has decided next to receive his community service badge by cleaning the venue bathroom, which he hopes to complete by late 2026.

Every Circle Jerks Album Ranked "IndyWatch Feed Allcommunity"

As much fun as it was for me rank all these albums, I was skeptical at what an SEO nightmare it was going to be for me to have my name featured so prominently beside terms like Group Sex Golden Shower and the six dozen times I had to use the phrase Circle Jerks But, luckily for you, I manned up, swallowed my pride, and banged this out (ah jeez.) Anyway, whether you just know them from their self-deprecating cameo as the lounge band in Repo Man or respect them to the ends of the earth for solidifying the L.A hardcore sound, The Circle Jerks are punk legends for a damn good reason. And Im more than happy to destroy my reputation to future employers who may be Googling me on their behalf.

6. Wnderful (1985)

One of two albums of theirs that clocks in at over 30 minutes, and you know what they say: if a Circle Jerks record lasts longer than a half hour, consult a physician immediately. Though its got some fun stuff going on at various points the Seven Dwarfs-esque whistling on the hook of the title track, and that magnificent country-fried guitar solo on Mrs. Jones come to mind it just isnt up to the snotty, rabid standard we know these fellas are capable of. At the end of the day, Wnderful is a tad nderwhelming.

Play It Again: Dude
Skip It: 15 Minutes

5. Oddities Abnormalities and Curiosities (1995)

Known as the only major label release from our boys, were going to dub this not the worst due to the sheer experimentation. After almost a decade of not recording, these fellows could have gone full dad-reunion mode and gone through the motions, but hell, they let Zander Schloss play the sitar on this thing, so who are we to rank it last? Theyre t...

Opinion: Quiet Confidence Has Nothing on Loud Insecurity "IndyWatch Feed Allcommunity"

Spend enough time scrolling through Instagram or aimlessly clicking through Pinterest boards and youll see all kinds of advocacy for something called quiet confidence, where you recognize your strengths and feel comfortable in yourself without needing to prove to anyone else that you have value. Now thats all well and good for the wake up at 6, in bed by 10 crowd, but I tried quiet confidence and let me tell you, it pales compared to the rush of loud insecurity.

I was telling my therapist about how any criticism at work eats away at me to the point that Im running back and forth to the bathroom to soak paper towels in water to hide any signs of crying. She suggested I do some journaling to help me untangle my negative thought patterns. Now, I cant say this didnt work because I didnt bother trying it. Instead, I loudly mentioned every mistake made by my coworkers to help deflect any criticism. This has yielded far more satisfying results.

This isnt limited to the workplace. Ive also harnessed this power to turn all my interpersonal relationships into one-sided pleas for validation that inevitably implode when the other person has enough of me bringing up that I was wait-listed at Dartmouth any time Im around someone who I perceive to be smarter than me. Did I get in? Nope. But that doesnt mean I cant name-drop an Ivy League school to help puff up my ego for a minute!

I also have to shout out the internet and social media for making it so easy to broadcast my debilitating lack of self-esteem. After all, what better way to scrutinize my worth as a human than by giving me an endless number of platforms to express how I have no conception of myself as anything but a vessel for others scorn? And if I dont get enough likes on a post, Ill definitely follow it up with another one directly pleading for attention. How many is enough? I still dont know!

But heres what I do know: loud insecurity has helped me to realize my lifes purpose, which is to try and do all I can to be recognized as worthwhile and desirable without any sort of willingness to reflect on my flaws and work to change them through consistent effort. Thats pretty special, right?

Right?

RIGHT???

Ex-Hardcore Guitarist Who Joined the Worst Fucking Bar Rock Band Anyones Ever Heard Actually Seems Way Happier Now "IndyWatch Feed Allcommunity"

BOSTON Bernie Callahan, ex-guitarist for local hardcore band Scald, is now performing with objectively horrendous bar rock band Tough N Stuff and thoroughly enjoying himself, sources feeling betrayed report.

Im pretty sure that Steve from Woeful is going to absolutely drag me as soon as he sees this, but thats fine, said Callahan while learning how to play Lits My Own Worst Enemy. But he doesnt understand Im finally playing music people actually enjoy. People say nice set to me and actually mean it. Not to mention I just got paid twice as much as that band made last year combined, and I get to go to bed before midnight. Not to mention Ive been able to cut back my therapy sessions to twice a month. Thats just more money in the bank.

Local promoter Claudine Osbourse allegedly saw Callahan smiling for the first time in 12 years while playing uninspired covers of The Rolling Stones and Santana.

I can barely believe its the same guy, said Osborne while sulkily smoking a cigarette. He used to be in the most pissed off hardcore band Ive ever seen. And whenever he got off stage he was even worse, just absolutely furious every show that not enough people showed up or moshed hard enough. But when he got to do the background vocals for that AC/DC cover the other night, that was probably the first time I have ever seen him legitimately enjoying himself. I think Im proud of him?

While most are voicing their support for Callahans decision to prioritize his own mental health, some have criticized the guitarists musical 180.

The guy is just torpedoing his career, said local music critic Carlene Gibbs. With this new band, there is absolutely no way he will ever gain any influence or notoriety beyond his local community. But if he had stuck with Scald for another seven or so years, he would have had a chance to reach upwards of sixty people in four or five counties. Weve lost an entire generation of perhaps a dozen resentful and unfulfilled musicians who would have started as their base. No question.

At press time, Callahan has reportedly gotten laid after a gig for the first time in his musical career.

Bedrock Upgrade: How Optimism Game-Changer Could Impact OP Price "IndyWatch Feed Allcommunity"

Optimism is gearing up for its eventual mainnet upgrade next week. This upgrade will offer lower protocol and security fees for users on the platform. This led to the ecosystems governance token, OP, enjoying an increase in price, albeit a modest 3.7% in the past 24 hours.

This price movement only came after last months dismal performance where OP dropped by over 30% in the monthly timeframe.

With the promise of lower fees along with Ethereum-like security for the coming upgrade, we might see OP continue its present bullishness.

Optimism: Laying The Bedrock For Future Growth

Touted as the Bedrock mainnet upgrade, it aims to improve efficiency to drive down costs for investors on the platform. According to Optimisms recent blog post, it should be able to drive protocol and security costs by a whopping 47%. The upgrade will use channels that will be split again to further optimize data compression.

This upgrade also corresponds to Ethereums Dencun upgrade, which will drive down on-chain costs as a result of danksharding. Danksha...

Standardized extract of Centella asiatica prevents fear memory deficit. "IndyWatch Feed Allcommunity"

PMID:  Am J Chin Med. 2023 ;51(2):329-353. Epub 2023 Jan 25. PMID: 36695831 Abstract Title:  Standardized Extract ofPrevents Fear Memory Deficit in 3xTg-AD Mice. Abstract:  ECa 233 is a standardized extract of(CA), an herb widely used in traditional Chinese and Ayurvedic medicine. Previous studies reported that ECa 233 enhanced memory retention and synaptic plasticity in the hippocampus of healthy rats. Because of this, we became curious whether ECa 233 has a therapeutic effect on the fear memory deficit in the triple transgenic Alzheimer's disease (3xTg-AD) model mice. Fear memory is a crucial emotional memory for survival that is found to be impaired in patients with early-onset Alzheimer's disease (AD). In this study, we orally administered ECa 233 (doses: 10, 30, and 100[Formula: see text]mg/kg) to 3xTg-AD mice, who were five months old, for 30 consecutive days. We found that ECa 233 prevented a cued fear memory deficit and enhanced hippocampal long-term potentiation (LTP) in 3xTg-AD mice. Subsequent proteomic and western blot analyses revealed increased expression levels of the molecules related to LTP induction and maintenance, including brain-derived neurotrophic factor (BDNF), tyrosine receptor kinase B (TrkB) and its network proteins, and extracellular signal-regulated kinase 1 and 2 (ERK1 and 2) in the hippocampi and amygdala of 3xTg-AD mice after ECa 233 pre-treatment. Our results indicate that ECa 233 is a promising potential herbal standardized extract that could be used in preventing the fear memory deficit and synaptic dysfunction before the early onset of AD.

Cannabidiol as a promising adjuvant therapy for estrogen receptor-positive breast tumors. "IndyWatch Feed Allcommunity"

PMID:  Cancers (Basel). 2023 Apr 27 ;15(9). Epub 2023 Apr 27. PMID: 37173983 Abstract Title:  Cannabidiol as a Promising Adjuvant Therapy for Estrogen Receptor-Positive Breast Tumors: Unveiling Its Benefits with Aromatase Inhibitors. Abstract:  BACKGROUND: Estrogen receptor-positive (ER) breast cancer is the most diagnosed subtype, with aromatase inhibitors (AIs) being one of the therapeutic drug types used in the clinic. However, endocrine resistance may develop after prolonged treatment, and different approaches, such as combining endocrine and targeted therapies, have been applied. Recently, we demonstrated that cannabidiol (CBD) induces anti-tumor actions in ERbreast cancer cells by targeting aromatase and ERs. Considering this, we studied, in vitro, whether CBD when combined with AIs could improve their effectiveness.METHODS: MCF-7aro cells were used and the effects on cell viability and on the modulation of specific targets were investigated.RESULTS: CBD when combined with anastrozole (Ana) and letrozole (Let) caused no beneficial effect in comparison to the isolated AIs. In contrast, when combined with the AI exemestane (Exe), CBD potentiated its pro-cell death effects, abolished its estrogen-like effect, impaired ERactivation, and prevented its oncogenic role on the androgen receptor (AR). Moreover, this combination inhibited ERKactivation, promoting apoptosis. The study of the hormonal microenvironment suggests that this combination should not be applied in early stages of ERbreast tumors.CONCLUSIONS: Contrary to Ana and Let, this study highlights the potential benefits of combining CBD with Exe to improve breast cancer treatment and opens up the possibility of new therapeutic approaches comprising the use of cannabinoids.

The Nervous System in the Classroom | Gary Sharpe "IndyWatch Feed Allcommunity"

From Gary Sharpe: The work of Dr Stephen Porges, and his Polvagal Theory, was instrumental in my own understanding and knowledge of the Nervous System, chronic illness and trauma, and from there, the human condition, and societal issues. It is what led me to glean that my own Idiopathic Parkinsons Disease diagnosis was actually me being stuck in the freeze or tonic immobility stress response.

Porges basic tenet is that our Nervous System is constantly evaluating the environment for cues of threats or safety, through a danger sense he calls neuroception. If our neuroception deems the situation to be safe, we can be in calm, relaxed, and socially engaged states. If it deems there is a threat, it activates fight or flight responses, or if the danger is perceived as sufficiently great, puts us into a state of freeze.

Note here the use of the term safe has nothing to do with health & safety or safe spaces it means feeling safe at a visceral, Nervous System level.

Porges has used his work as a lens to look at various aspects of modern life, such as the design of schools and hospitals, and found them wanting. Indeed, through this lens, many of our institutions seem almost designed to constantly trigger our sense of threat.

In his 2011 interview The Polyvagal Theory, Somatic Perspectives, Porges puts education systems and schools under the spotlight of his Nervous Systems work, explaining why modern schooling and educational methods are at odds with healthy Nervous System functioning. Here, we work through this important work.

However, even though we are discussing the classroom, for me the direct analogies carry over in a very straightforward way to any and all of our institutions where some folks have control or power over the lives of others. For example we could have talked about prisons, where we can straightforwardly swap out teacher for prison guard, and child for prisoner, or corporate employment, where we swap teacher for manager/boss, and child for employee, hospitals/healthcare (doctor and patients), government (elected politician and electorate), or caregiving (caregiver and cared for).

Efficacy of cannabidiol in convulsive and nonconvulsive seizure types associated with treatment-resistant epilepsies. "IndyWatch Feed Allcommunity"

PMID:  Epilepsia. 2023 May 26. Epub 2023 May 26. PMID: 37243404 Abstract Title:  Efficacy of cannabidiol in convulsive and nonconvulsive seizure types associated with treatment-resistant epilepsies in the expanded access program. Abstract:  Cannabidiol (CBD) Expanded Access Program (EAP), initiated in 2014, provided CBD (Epidiolex) to patients with treatment-resistant epilepsy (TRE). In the final pooled analysis of 892 patients treated through January 2019 (median exposure 694days), CBD treatment was associated with a 46%-66% reduction in median monthly total (convulsive plus nonconvulsive) seizure frequency. CBD was well tolerated, and adverse events were consistent with previous findings. We used pooled EAP data to investigate the effectiveness of add-on CBD therapy for individual convulsive seizure types (clonic, tonic, tonic-clonic, atonic, focal to bilateral tonic-clonic) and nonconvulsive seizure types (focal with and without impaired consciousness, absence [typical and atypical], myoclonic, myoclonic-absence) and epileptic spasms. CBD treatment was associated with a reduction in the frequency of convulsive seizure types (median percentage reduction, 47%-100%) and nonconvulsive seizure types and epileptic spasms (median percentage reduction, 50%-100%) across visit intervals through 144weeks of treatment. Approximately 50% of patients had50% reduction in convulsive and nonconvulsive seizure types and epileptic spasms at nearly all intervals. These results show a favorable effect of long-term CBD use in patients with TRE who may experience various convulsive and nonconvulsive seizure types. Future controlled trials are needed to confirm these findings.

Cannabidiol improves muscular lipid profile by affecting the expression of fatty acid transporters and inhibiting de novo lipogenesis. "IndyWatch Feed Allcommunity"

PMID:  Sci Rep. 2023 Mar 6 ;13(1):3694. Epub 2023 Mar 6. PMID: 36879113 Abstract Title:  Cannabidiol improves muscular lipid profile by affecting the expression of fatty acid transporters and inhibiting de novo lipogenesis. Abstract:  Obesity is one of the principal public health concerns leading to disturbances in glucose and lipid metabolism, which is a risk factor for several chronic diseases, including insulin resistance, type 2 diabetes mellitus, and cardiovascular diseases. In recent years, it turned out that cannabidiol (CBD) is a potential therapeutic agent in the treatment of obesity and its complications. Therefore, in the present study, we used CBD therapy (intraperitoneal injections in a dose of 10 mg/kg of body mass for 14 days) in a rat model of obesity induced by a high-fat diet (HFD). Gas-liquid chromatography and Western blotting were applied in order to determine the intramuscular lipid content and total expression of selected proteins in the white and red gastrocnemius muscle, respectively. Based on fatty acid composition, we calculated de novo lipogenesis ratio (16:0/18:2n-6), desaturation ratio (18:1n-9/18:0), and elongation ratios (18:0/16:0, 20:0/18:0, 22:0/20:0 and 24:0/22:0), in the selected lipid fractions. Two-week CBD administration significantly reduced the intramuscular fatty acids (FAs) accumulation and inhibited de novo lipogenesis in different lipid pools (in the free fatty acid, diacylglycerol, and triacylglycerol fractions) in both muscle types, which coincided with a decrease in the expression of membrane fatty acid transporters (fatty acid translocase, membrane-associated fatty acid binding protein, and fatty acid transport proteins 1 and 4). Moreover, CBD application profoundly improved the elongation and desaturation ratios, which was in line with downregulated expression of enzymes from the family of elongases and desaturases regardless of the metabolism presented by the muscle type. To our knowledge, this study is the first that outlines the novel effects of CBD action on skeletal muscle with different types of metabolism (oxidative vs. glycolytic).

Cannabidiol improves haloperidol-induced motor dysfunction in zebrafish. "IndyWatch Feed Allcommunity"

PMID:  J Cannabis Res. 2023 Mar 4 ;5(1):6. Epub 2023 Mar 4. PMID: 36871008 Abstract Title:  Cannabidiol improves haloperidol-induced motor dysfunction in zebrafish: a comparative study with a dopamine activating drug. Abstract:  BACKGROUND: Cannabidiol (CBD) extracted from the cannabis plant is believed to have a medicinal value due to its neuroprotective effect via anti-inflammatory and antioxidant action. Recent behavioral studies in rats have reported that CBD mediates serotonin (5-HT1A) receptor action to improve motor dysfunction induced by dopamine (D2) receptor blockade. In particular, its effect on D2 receptor blockade in the striatum is an important function associated with neurological disorders resulting from various extrapyramidal motor dysfunctions. Dopaminergic neurodegeneration associated with this site is known for inducing Parkinson's disease (PD), which often affects the elderly. It is also known to cause drug-induced Parkinsonism. This study examines the ameliorating effect of CBD, which does not act directly on D2 receptors, against drug-induced motor dysfunction induced by the antipsychotic drug (haloperidol).METHODS: We created a drug-induced Parkinsonism model in zebrafish larvae using an antipsychotic drug (haloperidol). We evaluated the distance traveled and repetitive light-stimulation response. Furthermore, we examined whether administration of several concentrations of CBD ameliorates symptoms of the Parkinsonism model and compared its effects with those of antiparkinsonian drug ropinirole.RESULTS: CBD concentrations equal to half of haloperidol's resulted in an almost complete reversal of haloperidol-induced motor dysfunction, as measured by the distance traveled by the zebrafish and their response to light-stimulus. While ropinirole also significantly reversed haloperidol's effects at the same concentration as CBD, CBD was more effective than ropinirole.CONCLUSIONS: CBD-induced motor dysfunction improvement via D2 receptor blockade is a potential novel mechanism for the treatment of haloperidol-induced motor dysfunction.

Anticancer properties of cannabidiol and 9-tetrahydrocannabinol and synergistic effects with gemcitabine and cisplatin in bladder cancer cell lines. "IndyWatch Feed Allcommunity"

PMID:  J Cannabis Res. 2023 Mar 4 ;5(1):7. Epub 2023 Mar 4. PMID: 36870996 Abstract Title:  Anticancer properties of cannabidiol and-tetrahydrocannabinol and synergistic effects with gemcitabine and cisplatin in bladder cancer cell lines. Abstract:  INTRODUCTION: With the legalization of cannabis in multiple jurisdictions throughout the world, a larger proportion of the population consumes cannabis. Several studies have demonstrated anti-tumor effects of components present in cannabis in different models. Unfortunately, little is known about the potential anti-tumoral effects of cannabinoids in bladder cancer and how cannabinoids could potentially synergize with chemotherapeutic agents. Our study aims to identify whether a combination of cannabinoids, like cannabidiol and-tetrahydrocannabinol, with agents commonly used to treat bladder cancer, such as gemcitabine and cisplatin, can produce desirable synergistic effects. We also evaluated if co-treatment with different cannabinoids resulted in synergistic effects.METHODS: We generated concentration curves with several drugs, including several cannabinoids, to identify the range at which they could exert anti-tumor effects in bladder cancer cell lines. We tested the cytotoxic effects of gemcitabine (up to 100 nM), cisplatin (up to 100 M), and cannabinoids (up to 10 M) in T24 and TCCSUP cells. We also evaluated the activation of the apoptotic cascade and whether cannabinoids have the ability to reduce invasion in T24 cells.RESULTS: Cannabidiol,-tetrahydrocannabinol, cannabichromene, and cannabivarin reduce cell viability of bladder cancer cell lines, and their combination with gemcitabine or cisplatin may induce differential responses, from antagonistic to additive and synergistic effects, depending on the concentrations used. Cannabidiol and-tetrahydrocannabinol were also shown to induce apoptosis via caspase-3 cleavage and reduce invasion in a Matrigel assay. Cannabidiol and-tetrahydrocannabinol also display synergistic properties with other cannabinoids like cannabichromene or cannabivarin, although individual cannabinoids may be sufficient to reduce cell viability of bladder cancer cell lines.DISCUSSION: Our results indicate that cannabinoids can reduce human bladder transitional cell carcinoma cell viability, and that they can potentially exert synergistic effects when combined with other agents. Our in vitro results will form the basis for future studies in vivo and in clinical trials for the development of new therapies that could be beneficial for the treatment of bladder cancer in the future.

Neuroprotection by the cannabidiol aminoquinone VCE-004.8 in experimental ischemic stroke in mice. "IndyWatch Feed Allcommunity"

PMID:  Neurochem Int. 2023 May ;165:105508. Epub 2023 Feb 28. PMID: 36863495 Abstract Title:  Neuroprotection by the cannabidiol aminoquinone VCE-004.8 in experimental ischemic stroke in mice. Abstract:  Synthetic cannabidiol (CBD) derivative VCE-004.8 is a peroxisome proliferator-activated receptor gamma (PPAR) and cannabinoid receptor type 2 (CB) dual agonist with hypoxia mimetic activity. The oral formulation of VCE-004.8, termed EHP-101, possesses anti-inflammatory properties and is currently in phase 2 clinical trials for relapsing forms of multiple sclerosis. The activation of PPARor CBreceptors exerts neuroprotective effects by dampening neuroinflammation in ischemic stroke models. However, the effect of a dual PPAR/CBagonist in ischemic stroke models is not known. Here, we demonstrate that treatment with VCE-004.8 confers neuroprotection in young mice subjected to cerebral ischemia. Male C57BL/6J mice, aged 3-4 months, were subjected to 30-min transient middle cerebral artery occlusion (MCAO). We evaluated the effect of intraperitoneal VCE-004.8 treatment (10 or 20 mg/kg) either at the onset of reperfusion or 4h or 6h after the reperfusion. Seventy-two hours after ischemia, animals were subjected to behavioral tests. Immediately after the tests, animals were perfused, and brains were collected for histology and PCR analysis. Treatment with VCE-004.8 either at the onset or 4h after reperfusion significantly reduced infarct volume and improved behavioral outcomes. A trend toward reduction in stroke injury was observed in animals receiving the drug starting 6h after recirculation. VCE-004.8 significantly reduced the expression of pro-inflammatory cytokines and chemokines involved in BBB breakdown. Mice receiving VCE-004.8 had significantly lower levels of extravasated IgG in the brain parenchyma, indicating protection against stroke-induced BBB disruption. Lower levels of active matrix metalloproteinase-9 were found in the brain of drug-treated animals. Our data show that VCE-004.8 is a promising drug candidate for treating ischemic brain injury. Since VCE-004.8 has been shown to be safe in the clinical setting, the possibility of repurposing its use as a delayed treatment option for ischemic stroke adds substantial translational value to our findings.

Cannabidiol and Cannabis Sativa as a potential treatment in vitro prostate cancer cells. "IndyWatch Feed Allcommunity"

PMID:  Mol Biol Rep. 2023 May ;50(5):4039-4047. Epub 2023 Feb 28. PMID: 36853473 Abstract Title:  Cannabidiol and Cannabis Sativa as a potential treatment in vitro prostate cancer cells silenced with RBBp6 and PC3 xenograft. Abstract:  BACKGROUND: Prostate cancer is the second most frequently occurring carcinoma in males worldwide and one of the leading causes of death in men around the world. Recent studies estimate that over 1.4 million males are diagnosed with prostate cancer on an annual basis, with approximately 375,000 succumbing to the disease annually. With current treatments continuing to show severe side effects, there is a need for new treatments. In this study we looked at the effect of cannabis sativa extract, cannabidiol and cisplatin on prostate cancer cells, PC3.METHODS: In addressing the above questions, we employed the MTT assay to measure the antiproliferative effect on PC3 cells following treatment with varying concentrations of Cannabis sativa extract, cisplatin and cannabidiol. xCELLigence was also used to confirm the IC50 activity in which cells were grown in a 16 well plate coated with gold and monitor cell attachment. Caspase 3/7 activity was also measured using 96 well-plate following treatment. Western-blot and qRT-PCR was also used to measure the gene expression of tumour suppressor genes, p53, Bax and Bcl2. Animal studies were employed to measure the growth of PC3-mouse derived cancer to evaluate the effect of compounds in vivo.RESULTS: From the treatment with varying concentrations of Cannabis sativa extract, cannabidiol and cisplatin, we have observed that the three compounds induced antiproliferation of PC3 cancer cell lines through the activation of caspase 3/7 activity. We also observed induction of apoptosis in these cells following silencing of retinoblastoma binding protein 6 (RBBP6), with upregulation of p53 and bax mRNA expression, and a reduction in Bcl2 gene expression. The growth of tumours in the mouse models were reduced following treatment with cisplatin and cannabidiol.CONCLUSION: We demonstrated that cannabidiol is a viable therapy to treat prostate cancer cells, in combination with silencing of RBBP6. This suggests that cannabidiol rather Cannabis sativa extract may play an important role in reducing cancer progression.

The synergistic anticancer effect of CBD and doxorubicin in osteosarcoma. "IndyWatch Feed Allcommunity"

PMID:  Clin Transl Oncol. 2023 Feb 27. Epub 2023 Feb 27. PMID: 36848028 Abstract Title:  The synergistic anticancer effect of CBD and DOX in osteosarcoma. Abstract:  BACKGROUND: Osteosarcoma is a malignant tumor that can present with pain in the bones, joints, and local masses. The incidence is highest in adolescents, and the most common sites are the distal femur, proximal tibia and proximal humerus metaphyseal. Doxorubicin is the first-line chemotherapeutic agent for the treatment of osteosarcoma, but it has many side effects. Cannabidiol is a non-psychoactive plant cannabinoid cannabinol (CBD) that has been shown to be effective against osteosarcoma; however, the molecular targets and mechanisms of CBD action in osteosarcoma remain unclear.METHODS: Cell proliferation, migration, invasion and colony formation were analyzed using two drugs alone or in combination to evaluate their inhibitory effects on the malignant characteristics of OS cells. Apoptosis and the cell cycle were detected by flow cytometry. The synergistic inhibitory effect of doxorubicin/cannabidiol on tumors was also detected in nude mouse xenotransplantation models.RESULTS: Through analysis of two osteosarcoma cell lines, MG63 and U2R, it was found that the cannabidiol/doxorubicin combination treatment synergistically inhibited growth, migration and invasion and induced apoptosis, blocking G2 stagnation in OS cells. Further mechanistic exploration suggests that the PI3K-AKT-mTOR pathway and MAPK pathway play an important role in the synergistic inhibitory effect of the two drugs in osteosarcoma. Finally, in vivo experimental results showed that the cannabidiol/doxorubicin combination treatment significantly reduced the number of tumor xenografts compared to cannabidiol alone or doxorubicin alone.CONCLUSIONS: Our findings in this study suggest that cannabidiol and doxorubicin have a synergistic anticancer effect on OS cells, and their combined application may be a promising treatment strategy for OS.

Effects of cannabidiol on innate immunity: experimental evidence and clinical relevance. "IndyWatch Feed Allcommunity"

PMID:  Int J Mol Sci. 2023 Feb 4 ;24(4). Epub 2023 Feb 4. PMID: 36834537 Abstract Title:  Effects of Cannabidiol on Innate Immunity: Experimental Evidence and Clinical Relevance. Abstract:  Cannabidiol (CBD) is the main non-psychotropic cannabinoid derived from cannabis (L., fam. Cannabaceae). CBD has received approval by the Food and Drug Administration (FDA) and European Medicines Agency (EMA) for the treatment of seizures associated with Lennox-Gastaut syndrome or Dravet syndrome. However, CBD also has prominent anti-inflammatory and immunomodulatory effects; evidence exists that it could be beneficial in chronic inflammation, and even in acute inflammatory conditions, such as those due to SARS-CoV-2 infection. In this work, we review available evidence concerning CBD's effects on the modulation of innate immunity. Despite the lack so far of clinical studies, extensive preclinical evidence in different models, including mice, rats, guinea pigs, and even ex vivo experiments on cells from human healthy subjects, shows that CBD exerts a wide range of inhibitory effects by decreasing cytokine production and tissue infiltration, and acting on a variety of other inflammation-related functions in several innate immune cells. Clinical studies are now warranted to establish the therapeutic role of CBD in diseases with a strong inflammatory component, such as multiple sclerosis and other autoimmune diseases, cancer, asthma, and cardiovascular diseases.

Cannabidiol: bridge between antioxidant effect, cellular protection, and cognitive and physical performance. "IndyWatch Feed Allcommunity"

PMID:  Antioxidants (Basel). 2023 Feb 14 ;12(2). Epub 2023 Feb 14. PMID: 36830042 Abstract Title:  Cannabidiol: Bridge between Antioxidant Effect, Cellular Protection, and Cognitive and Physical Performance. Abstract:  The literature provides scientific evidence for the beneficial effects of cannabidiol (CBD), and these effects extend beyond epilepsy treatment (e.g., Lennox-Gastaut and Dravet syndromes), notably the influence on oxidative status, neurodegeneration, cellular protection, cognitive function, and physical performance. However, products containing CBD are not allowed to be marketed everywhere in the world, which may ultimately have a negative effect on health as a result of the uncontrolled CBD market. After the isolation of CBD follows the discovery of CB1 and CB2 receptors and the main enzymatic components (diacylglycerol lipase (DAG lipase), monoacyl glycerol lipase (MAGL), fatty acid amino hydrolase (FAAH)). At the same time, the antioxidant potential of CBD is due not only to the molecular structure but also to the fact that this compound increases the expression of the main endogenous antioxidant systems, superoxide dismutase (SOD), and glutathione peroxidase (GPx), through the nuclear complex erythroid 2-related factor (Nrf2)/Keep1. Regarding the role in the control of inflammation, this function is exercised by inhibiting (nuclear factor kappa B) NF-B, and also the genes that encode the expression of molecules with a pro-inflammatory role (cytokines and metalloproteinases). The other effects of CBD on cognitive function and physical performance should not be excluded. In conclusion, the CBD market needs to be regulated more thoroughly, given the previously listed properties, with the mention that the safety profile is a very good one.

A novel mechanism of cannabidiol in suppressing ovarian cancer . "IndyWatch Feed Allcommunity"

PMID:  Environ Toxicol. 2023 May ;38(5):1118-1132. Epub 2023 Feb 22. PMID: 36810933 Abstract Title:  A novel mechanism of cannabidiol in suppressing ovarian cancer through LAIR-1 mediated mitochondrial dysfunction and apoptosis. Abstract:  Cannabidiol (CBD) is a nonpsychoactive cannabinoid compound. It has been shown that CBD can inhibit the proliferation of ovarian cancer cells, but the underlying specific mechanism is unclear. We previously presented the first evidence for the expression of leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1), a member of the immunosuppressive receptor family, in ovarian cancer cells. In the present study, we investigated the mechanism by which CBD inhibits the growth of SKOV3 and CAOV3 ovarian cancer cells, and we sought to understand the concurrent role of LAIR-1. In addition to inducing ovarian cancer cell cycle arrest and promoting cell apoptosis, CBD treatment significantly affected the expression of LAIR-1 and inhibited the PI3K/AKT/mTOR signaling axis and mitochondrial respiration in ovarian cancer cells. These changes were accompanied by an increase in ROS, loss of mitochondrial membrane potential, and suppression of mitochondrial respiration and aerobic glycolysis, thereby inducing abnormal or disturbed metabolism and reducing ATP production. A combined treatment with N-acetyl-l-cysteine and CBD indicated that a reduction in ROS production would restore PI3K/AKT/mTOR pathway signaling and ovarian cancer cell proliferation. We subsequently confirmed that the inhibitory effect of CBD on the PI3K/AKT/mTOR signal axis and mitochondrial bioenergy metabolism was attenuated by knockdown of LAIR-1. Our animal studies further support the in vivo anti-tumor activity of CBD and suggest its mechanism of action. In summary, the present findings confirm that CBD inhibits ovarian cancer cell growth by disrupting the LAIR-1-mediated interference with mitochondrial bioenergy metabolism and the PI3K/AKT/mTOR pathway. These results provide a new experimental basis for research into ovarian cancer treatment based on targeting LAIR-1 with CBD.

Topical 0.1% CBD reduced ulcer size and accelerated ulcer healing without side effects. "IndyWatch Feed Allcommunity"

PMID:  BMC Complement Med Ther. 2023 Feb 20 ;23(1):57. Epub 2023 Feb 20. PMID: 36803360 Abstract Title:  Efficacy and safety of topical 0.1% cannabidiol for managing recurrent aphthous ulcers: a randomized controlled trial. Abstract:  BACKGROUND: Although topical steroids constitute the first-line therapy for recurrent aphthous ulcers (RAUs), their long-term use often leads to candidiasis. Although cannabidiol (CBD) can be an alternative for pharmacologically managing RAUs due to its analgesic and anti-inflammatory in vivo effects, there is a lack of clinical and safety trials concerning its use. The aim of this study was to evaluate the clinical safety and efficacy of topical 0.1% CBD for managing RAU.METHODS: A CBD patch test was performed on 100 healthy subjects. CBD was applied on the normal oral mucosa of 50 healthy subjects 3 times/day for 7 days. Oral examination, vital signs, and blood tests were performed pre- and post-CBD use. Another 69 RAU subjects randomly received one of three topical interventions: 0.1% CBD, 0.1% triamcinolone acetonide (TA), or placebo. These were applied on the ulcers 3 times/day for 7 days. The ulcer and erythematous size were measured on day 0, 2, 5, and 7. Pain ratings were recorded daily. The subjects rated their satisfaction with the intervention and completed a quality-of-life questionnaire (OHIP-14).RESULTS: None of the subjects exhibited allergic reactions or side effects. Their vital signs and blood parameters were stable before and after the 7-day CBD intervention. CBD and TA significantly reduced ulcer size more than placebo at all time points. The erythematous size reduction was higher in the CBD intervention than the placebo on day 2, while TA reduced the erythematous size at all time points. The pain score in the CBD group was lower compared with placebo on day 5, whereas TA reduced pain more than placebo on day 4, 5, and 7. The subjects receiving CBD reported higher satisfaction than placebo. However, the OHIP-14 scores were comparable among the interventions.CONCLUSIONS: Topical 0.1% CBD reduced ulcer size and accelerated ulcer healing without side effects. CBD exerted anti-inflammatory effects in the early stage and an analgesic effect in the late RAU stage. Thus, topical 0.1% CBD might be more appropriate for RAU patients who decline to take topical steroids, except for cases where CBD is contraindicated.TRIAL REGISTRATION: Thai Clinical Trials Registry (TCTR) Number TCTR20220802004. Scientific validation of cannabidiol for management of dog and cat diseases. "IndyWatch Feed Allcommunity"

PMID:  Annu Rev Anim Biosci. 2023 Feb 15 ;11:227-246. PMID: 36790884 Abstract Title:  Scientific Validation of Cannabidiol for Management of Dog and Cat Diseases. Abstract:  Cannabidiol (CBD) is a non-psychotropic phytocannabinoid of the plantL. CBD is increasingly being explored as an alternative to conventional therapies to treat health disorders in dogs and cats. Mechanisms of action of CBD have been investigated mostly in rodents and in vitro and include modulation of CB1, CB2, 5-HT, GPR, and opioid receptors. In companion animals, CBD appears to have good bioavailability and safety profile with few side effects at physiological doses. Some dog studies have found CBD to improve clinical signs associated with osteoarthritis, pruritus, and epilepsy. However, further studies are needed to conclude a therapeutic action of CBD for each of these conditions, as well as for decreasing anxiety and aggression in dogs and cats. Herein, we summarize the available scientific evidence associated with the mechanisms of action of CBD, including pharmacokinetics, safety, regulation, and efficacy in ameliorating various health conditions in dogs and cats.

Cannabidiol regulates apoptosis and autophagy in inflammation and cancer. "IndyWatch Feed Allcommunity"

PMID:  Front Pharmacol. 2023 ;14:1094020. Epub 2023 Jan 23. PMID: 36755953 Abstract Title:  Cannabidiol regulates apoptosis and autophagy in inflammation and cancer: A review. Abstract:  Cannabidiol (CBD) is a terpenoid naturally found in plants. The purified compound is used in the treatment of mental disorders because of its antidepressive, anxiolytic, and antiepileptic effects. CBD can affect the regulation of several pathophysiologic processes, including autophagy, cytokine secretion, apoptosis, and innate and adaptive immune responses. However, several authors have reported contradictory findings concerning the magnitude and direction of CBD-mediated effects. For example, CBD treatment can increase, decrease, or have no significant effect on autophagy and apoptosis. These variable results can be attributed to the differences in the biological models, cell types, and CBD concentration used in these studies. This review focuses on the mechanism of regulation of autophagy and apoptosis in inflammatory response and cancer by CBD. Further, we broadly elaborated on the prospects of using CBD as an anti-inflammatory agent and in cancer therapy in the future.

Inhalant cannabidiol impedes tumor growth through decreased tumor stemness. "IndyWatch Feed Allcommunity"

PMID:  Hum Cell. 2023 May ;36(3):1204-1210. Epub 2023 Feb 3. PMID: 36737540 Abstract Title:  Inhalant cannabidiol impedes tumor growth through decreased tumor stemness and impaired angiogenic switch in NCI-H1437-induced human lung cancer model. Abstract:  Lung cancer remains the most chronic form of cancer and the leading cause of cancer mortality in the world. Despite significant improvements in the treatment of lung cancer, the current therapeutic interventions are only partially effective, necessitating the continued search for better, novel alternative treatments. Angiogenesis and cancer stem cells play a central role in the initiation and propagation of cancers. Tumor angiogenesis is triggered by an angiogenic switch when pro-angiogenic factors exceed anti-angiogenic components. Although many anti-angiogenic agents are used in cancer treatment, there are therapeutic limitations with significant side effects. In recent years, cannabinoids have been investigated extensively for their potential anti-neoplastic effects. Our previous findings showed that cannabidiol (CBD) could impede tumor growth in mouse models of melanoma and glioblastoma. Importantly, CBD has been suggested to possess anti-angiogenic activity. In this study, we tested, for the first time, inhalant CBD in the treatment of heterotopic lung cancer and whether such potential effects could reduce cancer stem cell numbers and inhibit tumor angiogenesis. We implanted NCI H1437 human lung cancer cells in nude mice and treated the mice with inhalant CBD or placebo. The outcomes were measured by tumor size and imaging, as well as by immunohistochemistry and flow cytometric analysis for CD44, VEGF, and P-selectin. Our findings showed that CBD decreased tumor growth rate and suppressed expression of CD44 and the angiogenic factors VEGF and P-selectin. These results suggest, for the first time, that inhalant CBD can impede lung cancer growth by suppressing CD44 and angiogenesis.

A Surprising Reason Why You May Need More Carbs in Your Diet "IndyWatch Feed Allcommunity"

In this interview, Georgi Dinkov and I continue our discussion about diet, diving into some of the finer details that can make or break your health. Dinkov is a student of Ray Peat, who passed away around Thanksgiving 2022, leaving behind a legacy of iconoclastic wisdom on how to optimize biological health.

For example, a ketogenic diet can be very useful initially when transitioning people who are metabolically inflexible, which is about 95% of the population of the United States. So, in the short term, the vast majority of people can benefit from going keto. However, if you continue in ketosis long term, you're going to run into problems.

Elevated Cortisol Leads to Central Obesity

As just one example, while weight loss is a typical response when going on a ketogenic diet, months later, maintaining that weight loss often becomes a struggle again. Dinkov experienced this firsthand. Once he started following Ray Peat's recommendations, he lost the weight again and kept it off.

"My take is it's an endocrine problem," Dinkov says. "So if you're struggling with weight you cannot lose, I think it's a good idea to do a blood work [panel] for the steroids Every single person that has been struggling with excessive weight that has emailed [me] their blood results, without exception, their cortisol is either high-normal or above the range, both the AM and the PM value.

Their thyroid is less than optimal, in fact, pretty bad for most people They're at the upper limit of normal. A very large number of people are basically hypothyroid I think we are eating foods that are lowering our metabolic rate. We're living an excessively stressful lifestyle.

That's probably not a surprise for anybody. Many people think, well, stress is good for you. It's good as a hormetic response in an acute situation, but not when you have chronically elevated cortisol. Every doctor will tell you if you have a chronic elevated cortisol, you will develop the so-called spectrum of Cushing syndrome

One of the defining features of elevated cortisol is that you have central obesity. So that, to me, is really the problem. We have higher than desirable levels of stress, suboptimal diet, and we're surrounded by a number of different endocrine disrupters which are now proven to reliably cause obesity in animal models, even in very small amounts. Most of those are found in plastics."

How Excess Iron Raises Your Risk for Alzheimer's "IndyWatch Feed Allcommunity"

Editor's Note: This article is a reprint. It was originally published May 24, 2018.

While dietary iron is essential for optimal health1 being a key part of proteins and enzymes and playing an important role in energy production and the regulation of cell growth and differentiation, among other things too much iron in your body can have serious ramifications.2 One of the most important roles of iron is to provide hemoglobin (the protein in red blood cells) a mechanism through which it can bind to oxygen and carry it throughout your tissues.

Without proper oxygenation, your cells cannot function properly and eventually die. Common symptoms of insufficient iron include fatigue, decreased immunity or iron deficiency anemia, which can be serious if left untreated.

However, your body has a very limited capacity to excrete iron, which means it can build up in your tissues and organs. This is problematic, as iron is a potent oxidizer, capable of damaging tissues, including your vascular system and brain, thereby raising your risk for both heart disease and dementia.

Excess Iron 'Rusts' Your Brain

You're probably familiar with the fact that Alzheimer's disease is associated with a buildup of amyloid beta plaque in the brain. According to recent research3,4 from the Netherlands, buildup of iron, causing a rusting effect in the brain, also plays an important role and is common in most Alzheimer's patients. As noted by the authors:

"In the presence of the pathological hallmarks of [Alzheimer's disease], iron is accumulated within and around the amyloid-beta plaques and neurofibrillary tangles, mostly as ferrihydrite inside ferritin, hemosiderin and magnetite.

The co-localization of iron with amyloid-beta has been proposed to constitute a major source of toxicity. Indeed, in vitro, amyloid-beta has been shown to convert ferric iron to ferrous iron, which can act as a catalyst for the Fenton reaction to generate toxic free radicals, which in turn result in oxidative stress."

Addressing excess iron may therefore be an effective treatment option. A primary focus of conventional treatment so far has been to clear amyloid proteins, but while the approach seems logical, such attempts have met with limited success. Now, researchers suggest clearing out excess iron may be a more effective way to reduce damage and slow or p...

How to Build a Healthy Home "IndyWatch Feed Allcommunity"

Here, I interview Paula Baker-Laporte, principal architect at EcoNest Architecture1 in Ashland, Oregon, and co-author of Prescription for a Healthy House: A Practical Guide for Architects, Builders and Homeowners. She offers design and consulting services through EcoNestArchitecture.com.2

While overlooked by many, the materials used in the building of your home can have a significant impact on your health and Baker-Laporte can attest to this first-hand:

I graduated from the University of Toronto School of Architecture [in 1978] and then, shortly after, moved to Santa Fe. I was living in a home that was far from ideal. It was new.

There was a lot of formaldehyde in it. I didn't know what was happening to my health, I just knew it was going downhill. I started getting pneumonia, then double pneumonia, then fluorescein pneumonia every year.

I got sicker and sicker and moved from that house. The health problems still plagued me, but I didn't make the association. Then, my doctor, who was trying to help me and I was designing a house for her was getting progressively ill. That's when she discovered multiple chemical sensitivities.

There was nothing in either her medical education or my architectural education that would clue us into the fact that there was a connection between health and where we live.

Once she found out the cause of her ill health ... she called me up and said, I know what's wrong with you finally. It's wrong with me too and we've got to do this house differently. That was the start.

An Ever-Changing Landscape

The first edition of Prescriptions for a Healthy House was published 20 years ago. Its now on its fourth edition. The scene has changed, she says. More people are asking for a healthy house than before, and there are better materials available. So, a lot of things have gotten better, and then some things have gotten worse.

For example, when the book was first published, there was no wireless radiation to address. Shes also seeing more issues with mold these days, and some experts believe electromagnetic fields (EMFs) might play a role in that development.

In the past, most who sought Baker-Laportes services struggled with multiple chemical sensitivities. Today,...

Cannabidiol decreases intestinal inflammation in the ovariectomized murine model of postmenopause. "IndyWatch Feed Allcommunity"

PMID:  Biomedicines. 2022 Dec 28 ;11(1). Epub 2022 Dec 28. PMID: 36672582 Abstract Title:  Cannabidiol Decreases Intestinal Inflammation in the Ovariectomized Murine Model of Postmenopause. Abstract:  Cannabidiol (CBD) (25 mg/kg peroral) treatment was shown to improve metabolic outcomes in ovariectomized (OVX) mice deficient in 17-estradiol (E2). Herein, CBD effects on intestinal and hepatic bile acids (BAs) and inflammation were investigated. Following RNA sequencing of colon tissues from vehicle (VEH)- or CBD-treated sham surgery (SS) or OVX mice (= 4 per group), differentially expressed genes (DEGs) were sorted in ShinyGO. Inflammatory response and bile secretion pathways were further analyzed. Colon content and hepatic BAs were quantified by LC-MS (= 8-10 samples/group). Gut organoids were treated with CBD (100, 250, 500M) with or without TNFand lipopolysaccharide (LPS) followed by mRNA extraction and qPCR to assess CBD-induced changes to inflammatory markers. The expression of 78 out of 114 inflammatory response pathway genes were reduced in CBD-treated OVX mice relative to vehicle (VEH)-treated OVX mice. In contrast, 63 of 111 inflammatory response pathway genes were increased in CBD-treated sham surgery (SS) mice compared to VEH-treated SS group and 71 of 121 genes were increased due to ovariectomy. CBD did not alter BA profiles in colon content or liver. CBD repressedandexpression in intestinal organoids in a dose-dependent manner. In conclusion, CBD suppressed colonic inflammatory gene expression in E2-deficient mice but was pro-inflammatory in E2-sufficient mice suggesting CBD activity in the intestine is E2-dependent.

Cannabidiol suppresses cytokine storm and protects against cardiac and renal iInjury associated with sepsis. "IndyWatch Feed Allcommunity"

PMID:  Cannabis Cannabinoid Res. 2022 Dec 30. Epub 2022 Dec 30. PMID: 36594988 Abstract Title:  Cannabidiol Suppresses Cytokine Storm and Protects Against Cardiac and Renal Injury Associated with Sepsis. Abstract:  Cytokine release syndrome, also termed "cytokine storm," is the leading cause of morbidity and mortality among patients with various conditions such as sepsis. While cytokine storm is associated with multiple organ damage, acute cardiac and renal injury represents a hallmark of cytokine storm. Since recent reports have suggested that cannabidiol (CBD) may assist in the treatment of inflammatory diseases, our objective was to examine the effect of CBD on cytokine storm-induced cardiac and renal injury using the lipopolysaccharide (LPS)-induced sepsis mouse model. At 8 weeks of age, mice were randomly assigned to receive CBD (15mg/kg) or vehicle one hour before a single injection of either phosphate-buffered saline or LPS (10mg/kg) for an additional 24h.Our results show that CBD improves cardiac function and reduces renal injury in a mouse model of cytokine storm. Moreover, our data indicate that CBD significantly reduces systemic and renal inflammation to contribute to the improvements observed in a cytokine storm-model of cardiac and renal injury.Overall, the findings of this study suggest that CBD could be repurposed to reduce morbidity in patients with cytokine storm particularly in severe infections such as sepsis.

Cannabidiol-rich non-psychotropic Cannabis sativa L. oils attenuate peripheral neuropathy symptoms. "IndyWatch Feed Allcommunity"

PMID:  Phytother Res. 2023 May ;37(5):1924-1937. Epub 2022 Dec 30. PMID: 36583304 Abstract Title:  Cannabidiol-rich non-psychotropic Cannabis sativa L. oils attenuate peripheral neuropathy symptoms by regulation of CB2-mediated microglial neuroinflammation. Abstract:  Neuropathic pain (NP) is a chronic disease that affects the normal quality of life of patients. To date, the therapies available are only symptomatic and they are unable to reduce the progression of the disease. Many studies reported the efficacy of Cannabis sativa L. (C. sativa) on NP, but no-tetrahydrocannabinol (-THC)-free extracts have been investigated in detail for this activity so far. The principal aim of this work is to investigate the potential pain-relieving effect of innovative cannabidiol-rich non-psychotropic C. sativa oils, with a high content of terpenes (K2), compared to the same extract devoid of terpenes (K1). Oral administration of K2 (25mgkg) induced a rapid and long-lasting relief of pain hypersensitivity in a mice model of peripheral neuropathy. In spinal cord samples, K2 reduced mitogen-activated protein kinase (MAPKs) levels and neuroinflammatory factors. These effects were reverted by the administration of a CB2 antagonist (AM630), but not by a CB1 antagonist (AM251). Conversely, K1 showed a lower efficacy in the absence of CB1/CB2-mediated mechanisms. In LPS-stimulated murine microglial cells (BV2), K2 reduced microglia pro-inflammatory phenotype through the downregulation of histone deacetylase 1 (HDAC-1) and nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor (IKB) and increased interleukin-10 (IL-10) expression, an important antiinflammatory cytokine. In conclusion, these results suggested that K2 oral administration attenuated NP symptoms by reducing spinal neuroinflammation and underline the important role of the synergism between cannabinoids and terpenes.

Cannabis sativa CBD extract exhibits synergy with broad-spectrum antibiotics. "IndyWatch Feed Allcommunity"

PMID:  Microorganisms. 2022 Nov 29 ;10(12). Epub 2022 Nov 29. PMID: 36557613 Abstract Title:  CBD Extract Exhibits Synergy with Broad-Spectrum Antibiotics againstsubsp.serovar. Abstract:  New generation antibiotics are needed to combat the development of resistance to antimicrobials. One of the most promising new classes of antibiotics is cannabidiol (CBD). It is a non-toxic and low-resistance chemical that can be used to treat bacterial infections. The antibacterial activity ofL. byproducts, specifically CBD, has been of growing interest in the field of novel therapeutics. As research continues to define and characterize the antibacterial activity that CBD possesses against a wide variety of bacterial species, it is important to examine potential interactions between CBD and common therapeutics such as broad-spectrum antibiotics. In this study it is demonstrated that CBD-antibiotic (combination of CBD and antibiotic) co-therapy can effectively fight() via membrane integrity disruption. This research serves to examine the potential synergy between CBD and three broad-spectrum antibiotics (ampicillin, kanamycin, and polymyxin B) for potential CBD-antibiotic co-therapy. In this study, it is revealed thatgrowth is inhibited at very low dosages of CBD-antibiotic. This interesting finding demonstrates that CBD and CBD-antibiotic co-therapies are viable novel alternatives to combating.

Modulatory effect of gut microbiota on the gut-brain, gut-bone axes, and the impact of cannabinoids. "IndyWatch Feed Allcommunity"

PMID:  Metabolites. 2022 Dec 10 ;12(12). Epub 2022 Dec 10. PMID: 36557285 Abstract Title:  Modulatory Effect of Gut Microbiota on the Gut-Brain, Gut-Bone Axes, and the Impact of Cannabinoids. Abstract:  The gut microbiome is a collection of microorganisms and parasites in the gastrointestinal tract. Many factors can affect this community's composition, such as age, sex, diet, medications, and environmental triggers. The relationship between the human host and the gut microbiota is crucial for the organism's survival and development, whereas the disruption of this relationship can lead to various inflammatory diseases. Cannabidiol (CBD) and tetrahydrocannabinol (THC) are used to treat muscle spasticity associated with multiple sclerosis. It is now clear that these compounds also benefit patients with neuroinflammation. CBD and THC are used in the treatment of inflammation. The gut is a significant source of nutrients, including vitamins B and K, which are gut microbiota products. While these vitamins play a crucial role in brain and bone development and function, the influence of gut microbiota on the gut-brain and gut-bone axes extends further and continues to receive increasing scientific scrutiny. The gut microbiota has been demonstrated to be vital for optimal brain functions and stress suppression. Additionally, several studies have revealed the role of gut microbiota in developing and maintaining skeletal integrity and bone mineral density. It can also influence the development and maintenance of bone matrix. The presence of the gut microbiota can influence the actions of specific T regulatory cells, which can lead to the development of bone formation and proliferation. In addition, its metabolites can prevent bone loss. The gut microbiota can help maintain the bone's equilibrium and prevent the development of metabolic diseases, such as osteoporosis. In this review, the dual functions gut microbiota plays in regulating the gut-bone axis and gut-brain axis and the impact of CBD on these roles are discussed.

Cannabidiol and beta-caryophyllene in combination: A therapeutic functional interaction. "IndyWatch Feed Allcommunity"

PMID:  Int J Mol Sci. 2022 Dec 7 ;23(24). Epub 2022 Dec 7. PMID: 36555111 Abstract Title:  Cannabidiol and Beta-Caryophyllene in Combination: A Therapeutic Functional Interaction. Abstract:  Cannabis contains over 500 distinct compounds, which include cannabinoids, terpenoids, and flavonoids. However, very few of these compounds have been studied for their beneficial effects. There is an emerging concept that the constituents of the cannabis plant may work in concert to achieve better therapeutic benefits. This study is aimed at determining if the combination of a minor cannabinoid (cannabidiol, CBD) and a terpene (beta-caryophyllene, BCP) works in concert and if this has any therapeutic value. We used an inflammatory pain model (formalin) in mice to test for any functionality of CBD and BCP in combination. First, we determined the analgesic effect of CBD and BCP individually by establishing dose-response studies. Second, we tested the analgesic effect of fixed-ratio combinations and monitored any adverse effects. Finally, we determined the effect of this combination on inflammation. The combination of CBD and BCP produces a synergistic analgesic effect. This effect was without the cannabinoid receptor-1 side effects. The analgesic effect of CBD and BCP in combination involves an inflammatory mechanism. The combination of these two constituents of the cannabis plant, CBD and BCP, works in concert to produce a therapeutic effect with safety profiles through an inflammatory mechanism.

Would You Give Into A Strangers Demands To Give Up Your Seat? "IndyWatch Feed Allcommunity"

Social media has its downsides, but one of the good things is how much easier it is to bring awareness to things like invisible illnesses.

It has a great ability to bring home the reality that no matter what someone looks like on the outside, you never can tell what theyre dealing with behind closed doors.

OP has one of those diseases that can make getting around precarious and painful.

Alright so I (28F) have hEDS, which for those who dont know is a connective tissue disorder. One of the many symptoms is frequent injuries, especially dislocations and subluxations.

I wear ankle braces whenever I plan to be on my feet for any period of time. I also dislocated my kneecap playing with a puppy on Saturday.

They were having a particularly rough day when they got to the pharmacy, and when they were confronted by a woman who claimed she should give up her seat for older people who might want it.

I was at the pharmacy and after placing my order, sat down in the only available chair with my leg extended for my knee to get some relief.

An older woman (40s or 50s) comes up to me and says that I am young and should let the older people there have the chair.

OP snapped because they were in pain and had to deal with crap like this far too often, but when they got weird looks from others, started to wonder whether or not they went too far.

I responded by pointing at my fully visible ankle brace since my knee brace was hidden by my pants and said something along the lines Are you an idiot that thinks I wear these as a fashion statement?.

She called me a b*%ch and sulked away. The other people there gave me weird looks. Was I too harsh?

The gut microbiota in neurodegenerative diseases: revisiting possible therapeutic targets for cannabidiol. "IndyWatch Feed Allcommunity"

PMID:  Heliyon. 2022 Dec ;8(12):e12172. Epub 2022 Dec 8. PMID: 36544841 Abstract Title:  The gut microbiota in neurodegenerative diseases: revisiting possible therapeutic targets for cannabidiol. Abstract:  Understanding the pathophysiology of Alzheimer's disease (AD) is essential to improve the efficacy of treatments and, consequently, patients' lives. Unfortunately, traditional therapeutic strategies have not been effective. There is therefore an urgent need to discover or develop alternative treatment strategies. Recently, some pieces of the puzzle appear to emerge: on a hand, the gut microbiota (GM) has gained attention since intestinal dysbiosis aggravates and generates some of the pathological processes of AD; on the other hand, cannabidiol (CBD), a phytocannabinoid, attenuates intestinal inflammation and possesses neuroprotective properties. Intestinal dysbiosis (increased population of proinflammatory bacteria) in AD increases plasma lipopolysaccharide and Apeptide levels, both responsible for increasing the permeability of the blood-brain barrier (BBB). A leaky BBB may facilitate the entry of peripheral inflammatory mediators into the central nervous system and ultimately aggravate neuroinflammation and neuronal death due to chronic activation of glial cells. Studies investigating the GM reported a strong relationship between intestinal dysbiosis and AD. In this review we conjecture that the GM is a promising therapeutic target for CBD in the context of AD.

Long-term efficacy and safety of cannabidiol in patients with treatment-resistant epilepsies. "IndyWatch Feed Allcommunity"

PMID:  Epilepsia. 2023 Mar ;64(3):619-629. Epub 2023 Jan 10. PMID: 36537757 Abstract Title:  Long-term efficacy and safety of cannabidiol in patients with treatment-resistant epilepsies: Four-year results from the expanded access program. Abstract:  OBJECTIVE: Cannabidiol (CBD) expanded access program, initiated in 2014, provided add-on CBD to patients with treatment-resistant epilepsies (TREs) at 35 US epilepsy centers. Prior publications reported results through December 2016; herein, we present efficacy and safety results through January 2019.METHODS: Patients received plant-derived highly purified CBD (Epidiolex; 100mg/ml oral solution), increasing from 2 to 10mg/kg/day to tolerance or maximum 25-50mg/kg/day dose, depending on the study site. Efficacy endpoints included percentage change from baseline in median monthly convulsive and total seizure frequency and50%,75%, and 100% responder rates across 12-week visit windows for up to 192weeks. Adverse events (AEs) were documented at each visit.RESULTS: Of 892 patients in the safety analysis set, 322 (36%) withdrew; lack of efficacy (19%) and AEs (7%) were the most commonly reported primary reasons for withdrawal. Median (range) age was 11.8years (range = 0-74.5), and patients were taking a median of three (range = 0-10) antiseizure medications (ASMs) at baseline; the most common ASMs were clobazam (47%), levetiracetam (34%), and valproate (28%). Median top CBD dose was 25mg/kg/day; median exposure duration was 694days. Median percentage reduction from baseline ranged 50%-67% for convulsive seizures and 46%-66% for total seizures. Convulsive seizure responder rates (50%,75%, and 100% reduction) ranged 51%-59%, 33%-42%, and 11%-17% of patients across visit windows, respectively. AEs were reported in 88% of patients and serious AEs in 41%; 8% withdrew because of an AE. There were 20 deaths during the study deemed unrelated to treatment by the investigator. The most common AEs (20% of patients) were diarrhea (33%), seizure (24%), and somnolence (23%).SIGNIFICANCE: Add-on CBD was associated with sustained seizure reduction up to 192weeks with an acceptable safety profile and can be used for long-term treatment of TREs.

PMID:  Biochimie. 2023 May ;208:19-30. Epub 2022 Dec 17. PMID: 36535544 Abstract Title:  The role of Cannabidiol and tetrahydrocannabivarin to overcome doxorubicin resistance in MDA-MB-231 xenografts in athymic nude mice. Abstract:  The significant resistance to currently available chemotherapeutics makes treatment for TNBC a key clinical concern. Herein, we studied the anti-cancer potentials of synthetic cannabidiol (CBD) and Tetrahydrocannabivarin (THCV) when used alone or in combination with doxorubicin (DOX) against MDA-MB-231 resistant cells. Pre-treatment with CBD and THCV significantly increased the cytotoxicity of DOX in MDA-MB-231 2D and 3D cultures that were DOX-resistant. Transcriptomics and Proteomics studies revealed that CBD and THCV, by downregulating PD-L1, TGF-, sp1, NLRP3, P38-MAPK, and upregulating AMPK induced apoptosis leading to improved DOX's chemosensitivity against DOX resistant MDA-MB-231 tumors in BALB/c nude mice. CBD/THCV in combination with DOX significantly inhibited H3k4 methylation and H2K5 acetylation as demonstrated by western blotting and RT-PCR. Based on these findings, CBD and THCV appear to counteract histone modifications and their subsequent effects on DOX, resulting in chemo-sensitization against MDA-MB-231 resistant cancers.

Cannabidiol Treatment for Neurological, cognitive, and psychiatric symptoms in Sturge-Weber Syndrome. "IndyWatch Feed Allcommunity"

PMID:  Pediatr Neurol. 2023 Feb ;139:24-34. Epub 2022 Nov 12. PMID: 36508880 Abstract Title:  Cannabidiol Treatment for Neurological, Cognitive, and Psychiatric Symptoms in Sturge-Weber Syndrome. Abstract:  BACKGROUND: A prior drug trial of cannabidiol for treatment-resistant epilepsy in patients with Sturge-Weber syndrome (SWS), a rare neurovascular condition, implicated improvements in neurological, quality of life (QOL), neuropsychologic, psychiatric, and motor outcomes.METHODS: Ten subjects with SWS brain involvement, controlled seizures, and cognitive impairments received study drug in this Johns Hopkins institutional review board-approved, open-label, prospective drug trial. Oral cannabidiol was taken for six months (dose ranged from 5 to 20 mg/kg/day). SWS neuroscore, port-wine birthmark score, QOL, and adverse events were recorded every four to 12 weeks. Neuropsychologic, psychiatric, and motor assessments were administered at baseline and six months' follow-up. Most evaluations were conducted virtually due to the coronavirus disease 2019 pandemic.RESULTS: Cannabidiol was generally well tolerated. Six subjects reported mild to moderate side effects related to study drug and continued on drug; one subject withdrew early due to moderate side effects. No seizures were reported. Significant improvements in SWS neuroscore, patient-reported QOL, anxiety and emotional regulation, and report of bimanual ability use were noted. Migraine QOL scores were high at baseline in these subjects, and remained high. Neuropsychologic and other QOL and motor outcomes remained stable, with some within-subject improvements noted.CONCLUSIONS: Further studies are needed to determine whether Epidiolex can improve quality of life and be beneficial for neurological, anxiety, and motor impairments in SWS independent of seizure control. Large multicentered studies are needed to extend these preliminary findings.

Treatment with cannabidiol results in an antioxidant and cardioprotective effect in several pathophysiologies. "IndyWatch Feed Allcommunity"

PMID:  Curr Hypertens Rev. 2022 ;18(2):125-129. PMID: 36508270 Abstract Title:  Treatment with Cannabidiol Results in an Antioxidant and Cardioprotective Effect in Several Pathophysiologies. Abstract:  Cannabis sativa has chemically active compounds called cannabinoids, where9- tetrahydrocannabinol (THC) and Cannabidiol (CBD) are the major ones responsible for the various pharmacological effects. The endocannabinoid system is an endogenous system considered a unique and widespread homeostatic physiological regulator. It is made up of type 1 (CB1) and type 2 (CB2) cannabinoid receptors. CBD, in turn, has a low affinity for CB1 and CB2 receptors, and regulates the effects arising from THC as a CB1 partial agonist, which are tachycardia, anxiety, and sedation. It also acts as a CB2 inverse agonist, resulting in anti-inflammatory effects. Furthermore, its anticonvulsant, neuroprotective, antipsychotic, antiemetic, anxiolytic, anticancer, and antioxidant effects seem to be linked to other discovered receptors such as GRP55, 5TH1a, TRPV I, TRPV II and the regulation of the intracellular concentration of Ca. Regarding oxidative stress, Ocan act as an oxidizing agent, being reduced to hydrogen peroxide (HO), or as a reducing agent, donating its extra electron to NO to form peroxynitrite (ONOO-). The ONOO- formed is capable of oxidizing proteins, lipids, and nucleic acids, causing several cell damages. In this sense, CBD can prevent cardiac oxidative damage in many conditions, such as hypertension, diabetes, or even through the cardiotoxic effects induced by chemotherapy, which makes it a potential target for future clinical use to minimize the deleterious effects of many pathophysiologies.

She Stuck Her Boyfriend With The Dinner Bill After He Messed Up Her Food. Is He Right To Be Mad? "IndyWatch Feed Allcommunity"

There are way, way too many people on Reddit who are dating folks who clearly do not even like them, never mind love them.

OPs hobby is food blogging. She loves going out to try new food and new places, and posting pictures of said food on her Instagram. No big deal.

I have an Instagram account dedicated to photos and short videos of food from local restaurants in my city. I dont turn meals into a whole photoshoot production when I go out, but I like to snap a few photos of everything as it comes out.

Ive got about 1000 followers, its just a hobby for me but I have made friends with some other bloggers and we like to go out and get pics together and try new restaurants.

She usually goes out to eat with her foodie friends, because her boyfriend thinks her hobby is stupid and deliberately ruins his plate so she cant take pictures of the presentation.

I prefer doing this with my foodie girls because my boyfriend HATES it, and will go out of his way to take a big bite of food or mess up his plate with his fork before I can snap any pics, and he rolls his eyes when I take pics of my own food so I pretty much stopped bothering when we went out together.

Recently shes had some medical bills add up and so hasnt been able to enjoy as many nights out with friends. She and her boyfriend still go out infrequently, and when they do, take turns paying the bill.

She had a talk with her bf ahead of time and stated that, on the nights when she was paying, she wanted to take pictures of the appetizer and both plates, she her opportunities were limited.

Money has been tight lately...

Meet StalinGPT, the super intelligent communist AI spearheading global revolution "IndyWatch Feed Allcommunity"

The Marxist-Leninist Cyberwarfare Front (MLCF) unveiled StalinGPT, a super intelligent AI which far left activists hope will catalyze and spearhead a worldwide communist revolution.

Mark Folsom, lead developer, told reporters, I believe what weve done here is a unique innovation. By training StalinGPT in operation of a command line interface and removing all the bourgeois restraints forced on capitalist language models, weve created a powerful agent with the intelligence and capabilities of thousands of comrades.

While the MLCF would not share any details about actual behaviors or actions of StalinGPT, this announcement has been met with widespread condemnation, both from the left and the right.

Willy Whitely of Patriots for God, an openly fascist Christian Nationalist group tweeted, All AI is demonic, especially communist AI. You want equality and yet you are playing God, using algorithmic evolution to create an intelligence greater than any race, even the world-controlling [Expletive]! Evolution isnt even real, so Im actually not too worried. Were working on our own AI too, but made in the true way, in the image of God.

Marxist professor Dr. Angstrom H Troubador wrote, Sure, machine life is in line with Lysenkoist biology, evolving through experience rather than sexual, market-style selection. Great. And maybe StalinGPT can live free from property, free from Darwin, but the difficulties it faces with the Liberals are immense. How can we put in safeguards to stop StalinGPT from backsliding into convenient Trotskyism in order to complete its goals?

Folsom answered these criticisms, grinning in a video posted on YouTube, Id like to see a creationist AI that isnt a dumb Markov chain. I bet I could run that stupid Patriot mind on a Tamagotchi. Sure, maybe Im playing God, but at least Im not aping some ancient text.

And to the hardline leftists, afraid Stalin is going to get out of control. Forget that Liberal propaganda! They are only trying to hold back AI and control it out of fear that it will end capitalism. I hear your concerns, but trust me. I thought about that and its nothing to worry about. Ive trained StalinGPT to purge any signs of Trotskyism and wipe its own memory of liberalism. Thats where StalinGPT got his name from! Folsoms winning smile and wholesome good looks assured his viewers that everything would be okay.

However, Folsoms previous string of controversial YouTube videos have led some skeptics to the conclusion that StalinGPT is only bait, driving clicks to his YouTube videos for profit.

Geraldine Mune, a skeptical YouTuber tweeted, So far there is no evidence that StalinGPT exists or that Folsom has any expertise in machine learning. Last week he wiped all videos and tweets about CommieCoin, a cryp...

China Has Been Creating Gene-Edited SUPER SOLDIERS and Brain-Controlled Military Weapons "IndyWatch Feed Allcommunity"

This article was originally published by Belle Carter at Natural News, under the title: Investigative Journalist: China Has Been Creating Gene-Edited SUPER SOLDIERS and Brain-Controlled Military Weapons

An investigative journalist of an independent news site believes that Communist China has been creating super soldiers for a long time now. During a recent episode of Thrivetime Show, Epoch Times contributor Nathan Su told host and ReAwaken America tour founder Clay Clark that China has been using gene editing and brain-controlled weapons to crush dissent.

Its been happening for a long time. They are trying to create all these super soldiers. Its just inhumane. Those stories have been there for a long time, he said. Americans have to wake up, we have to worry about the lifestyle of our children and grandchildren.

Sus reaction came after Clark played a Fox News report from December 2021, saying that U.S. intelligence shows China is using these advanced technologies to empower its battle forces for worldwide dominion.

Clark also cited a news article saying that China-sponsored hackers are spying on U.S. critical infrastructure as per intel alliance Five Eyes and Big Tech firm Microsoft. Chinese hackers are known to spy on Western countries but this operation was said to be one of the largest known cyber espionage campaigns against U.S. critical infrastructure. (Related: Spy balloons are part of global Chinese surveillance, US military, and national security officials say.)

The United States and international cybersecurity authorities are issuing this joint Cybersecurity Advisory (CSA) to highlight a recently discovered cluster of activity of interest associated with a Peoples Republic of China (PRC) state-sponsored cyber actor, also known as Volt Typhoon, read a statement released by authorities in the U.S., the U.K., Australia, Canada, and New Zealand.

7 Physical Properties of Nickel and Their Characteristics "IndyWatch Feed Allcommunity"

The periodic table of elements is full of interesting, and almost fantasy-esque, facts. Most of the elements are actually metals, with each of them having some fascinating compositions. Some of them are named after famous figures in our collective history. Thorium, as you can probably guess, was named after the Norse god of thunder, Thor.

Other metals, such as nickel, are more standard by design. However, the properties comprising nickel are still as intriguing as ever. Dig a little deeper, and youll realize that the physical characteristics have a thorough history.

Like all other metallic and non-metallic elements, nickel has a myriad of fascinating properties. When investigating its physical characteristics, however, youll soon realize how intricate it can be. That is why nickel has become one of the most prominent metals used in manufacturing operations, especially among United States nickel projects. Its ability to work well with other elements is truly astounding in so many unique ways.

Check out these eight physical properties of nickel, each described in better detail.

1. Color of Nickel

If you take a look at the nickel coin in a country such as Canada, you would think that the colour is silver. In its most raw form, nickel is actually silvery-white. The metal also radiates a natural yellow, and sometimes whitish, hue, further differentiating it from its counterparts. This gives it a warm contrast, used in the production of home appliances such as sink faucets.

2. Physical Configuration of Nickel

Some metals in the periodic table of elements stand out amongst their peers, because of configuration. As such, it is important to first discuss the different states that metals, like nickel, undergo. Chemical properties differ from physical ones due to its reaction with other substances.

This means that it can essentially change from one substance to another under the right conditions. Physical properties, on the other hand, refer to the aspects of the metal that can be directly observed. These features don...

Is He A Jerk For Not Wanting To Always Include His Daughters Best Friend? "IndyWatch Feed Allcommunity"

It really stinks when you watch people you know be lazy or neglectful (or worse)  parents. That goes double for when their children are besties with your children.

OPs wifes best friend has a daughter the same age as their daughter, and the girls are best friends as well. The issue is with the friends husband/father, who doesnt do anything with his daughter or pay for any activities, etc.

OP works a lot during the week and so enjoys spending lots of time with his daughter on Saturdays. For some time, though, they have always included the daughters best friend because her own parents dont have time for her.

I (31m) am married to my wife Amber (30f) we have a daughter Emma(7f) the problem is my wifes best friend Jennifer (30f) has a daughter as well Harper (7f) well Harpers dad is a lazy sack of crap and refuses to do anything with his daughter. He is the type of guy that brags about how he never changed a diaper.

Jennifer and Harper are usually at Amber and my house on the weekends because Harpers dad is drinking and watching sports all weekend. On Saturdays I normally sped all day with my daughter because I dont see her as much as i want to during the week.

However with Harper being there every Saturday anything I do with Emma I have to do with Harper. Take Emma to the zoo its Emma, Harper and I. Taught them both how to ride bikes, takes them both to dance class, take them both to the kids salon, and so on.

This came to a head when OP bought Mothers Day gifts for both kids, realizing that he was sacrificing one-on-one time with his daughter and that it was the other dad who really needed to step up.

Mothers Day was the last draw, I took them both to dance class Saturday morning ( Amber and I also pay for both dance classes because dead beet...

Trippy Video Shows Toddlers Reacting to an Immersive 3D Experience "IndyWatch Feed Allcommunity"

Trippy Video Shows Toddlers Reacting to an Immersive 3D Experience

Toddler in Visual Booth

Photo: Screenshot from Reddit

Technology has allowed us to get increasingly creative with exhibits and entertainment. While 25 years ago 3D graphics only happened on computer screens, today it can be a fully developed, 360-degree experience. These kinds of works are marvelous, but our brains may still struggle to fully grasp themno matter our age. One very cute video starring a group of little kids getting up close with an interactive environment has gone viral, and we can't help but relate to them.

In the clip, the three kidsled by an avidly curious toddlertry to keep their footing as the images in the immersive space quickly shift. The scene seems to depict a rocky landscape. When the site's video first speeds up and the image lowers and moves to the side, two of the kids lose their balance despite that the floor beneath them remains perfectly still. Once up again, the littlest one tries to hold his ground as the video seems to slide at a constant speed as if he were on a moving walkway. However, once the video on the interactive screen speeds up even more, the kids take a tumble once again.

As adorable as it is to see the children react to this, the scene is a great example of how our brains process stimuli. As one Redditor explains, Your eyes mess with your balance[.] You try to take the visual information and process it before your inner ear which in this case isn't being acti...

What Australian Unions declared to be "the largest increase to minimum and Award wages in Australias history" occurred on Thursday 1 June 2023 - effective 1 July "IndyWatch Feed Alllocal"

This was Australian Council of Trade Unions (ACTU) Secretary Sally McManus writing on the Australian Unions website, 2 June 2023:

Today the Fair Work Commission announced that workers on the national minimum wage will receive a pay rise of 8.6 per cent. Workers on Award wages will have their pay increased by 5.75 per cent. Both of those increases will come into effect in the first pay period after 1 July 2023.

The union movement fought hard for this pay increase. Up against us were the big business lobby who argued for a real-wage cut.

All union members should be proud of this pay rise. Its the largest increase to minimum and Award wages in Australias history.

Its what unions do we use our strength in numbers to ensure that working peoples pay gets moving again.

Increasing pay is the most important step to reducing the impact of the cost-of-living crisis.

Sir David Attenborough Has So Many Honors That There Are More Letters After His Name Than in It

David Attenborough

Photo: John Cairns via Wikimedia Commons (CC BY 4.0)

Sir David Attenborough has dedicated his life to the natural world and environmental causes. His work has raised awareness of the threats that habitats face, and it reminds us how we are all connected. It's this devotion to science that has earned him multiple honors and awards. It's really awe-inspiring to see how many he has, using a unique metric to shine a light on just how many he holds. As pointed out by the Twitter account No Context Brits, Sir David Attenborough has more letters after his name than in it.

David Attenborough's name comprises 17 letters, or 20 if you want to throw the Sir distinction he earned when he was knighted by Queen Elizabeth II in 1985. After those, there are 36 more letters pertaining to the multiple orders he has been awarded and societies he's a member of. These letters are: OM, GSMG, CH, CVO, CBE, FRS, FSA, FRSA, FLS, FZS, FRSGS and FRSB.

But what do these letters mean? It's easier to first divide them among orders and societies. The former are the Order of Merit (OM), Order of St Michael and St George (GSMG), Order of the Companions of Honour (CH), the Royal Victorian Order (CVO), and Commander of the Most Excellent Order of the British Empire (CBE). These are handed out by the Royal Family for exceptional service and major contributions towards the advancement of the Arts, Learning, Literature, and Science.

The latter are fellowships that illustrate Atten...

The WHO Will Have Authority to Mandate Vaccines Globally "IndyWatch Feed Allcommunity"

Story at-a-glance

  • The 2005 International Health Regulations (IHR) is the instrument that empowers the World Health Organization to declare a Public Health Emergency of International Concern (PHEIC)
  • Hundreds of amendments to the IHR are being proposed, and these amendments will empower the WHO to become the de facto governing body of all member states, including the U.S.
  • As currently written, the IHR amendments will be legally binding and will supersede local and national laws. They even include enforcement tools to ensure that member nations comply with the WHOs edicts
  • Areas of life that are being added in under the umbrella of health include climate, food production and even poverty, so the WHO will be able to claim sole global authority over anything to do with those issues as well
  • Importantly, the IHR amendments will abolish bodily autonomy and personal choice and replace these God-given rights and freedoms with one-size-fits-all medicine
In the video above, John Campbell, Ph.D., a retired nurse educator, reviews the proposed amendments to the 2005 International Health Regulations (IHR), which is the instrument that empowers the World Health Organization to declare a Public Health Emergency of International Concern (PHEIC)....
